Dual-Selective Magnetic Analysis of Extracellular Vesicle Glycans

Abstract: Extracellular vesicles (EVs) are emerging circulating biomarkers and contain diverse sugar modifications (glycans) that can mediate disease progression. The analysis of EV glycans, however, is challenging due to technology limitations. We have developed a dedicated platform to directly profile EV glycans in native biofluids. The all-magnetic technology utilizes rationally designed nanoparticles and integrated sensors to specifically measure EV-bound glycans but not that of free-floating proteins. The assay is … Show more

“…Using STARs functionalized with antibodies, we developed the assay to achieve direct glycan profiling of cancer-associated EVs in patient specimens. Specifically, we prepared STARs with antibodies against CD24 34 , 35 , a known cancer antigen, to enrich and measure putative tumor-derived EVs. Multiplexed glycan profiling showed highly varied glycan signatures among the clinical samples (Fig.…”

“…As demonstrated in various cancer types, the discovery of prognostic gene signatures could provide clinicians with tools for better disease management (52)(53)(54). The potential utility of unique signatures on extracellular vesicles and glycans could also be harnessed for patient prognostication (55). Minimally invasive surgery options in the background of recurrent ovarian cancer treated with HIPEC have recently been suggested as a means of reducing surgical risk and post-operative complications in the subgroup of patients indicated for such surgery, providing desirable outcomes with a reported upside of increased peritoneal uptake of HIPEC (56,57).…”

“…Interestingly, a new integrated analytical platform, termed the integrated magnetic analysis of glycans in extracellular vesicles (iMAGE), was developed to directly analyze the EV glycosylation profile in biological samples. This platform aims to facilitate the EV glycome analysis taking advantage of the magnetic nanotechnologies [ 218 ]. The effectiveness of this strategy was evaluated by spiking kidney and brain cancer-EVs into urine and serum EV-depleted samples, respectively, and analyzing the glycan signatures.…”

“…The distinction of these patients was possible by the increased signal of different lectins associated with a poor prognosis, including the Jacalin, ConA, RCA120, PHA-E, STA, LEL, WGA, DSL and LCA lectins. Although further studies are needed to prove the iMAGE platform’s robustness, this new method of profiling glycans may prove to be very useful in the search for novel biomarkers in cancer research [ 218 ]. Although several techniques can be used to analyze glycans, their study faces several technical challenges.…”
