Dual window method for processing spectroscopic optical coherence tomography signals with simultaneously high spectral and temporal resolution

Abstract: Current methods for analysis of spectroscopic optical coherence tomography (SOCT) signals suffer from an inherent tradeoff between time (depth) and frequency (wavelength) resolution. Here, we present a dual window (DW) method for reconstructing time frequency distributions (TFDs) that applies two orthogonal Gaussian windows that independently determine the spectral and temporal resolution. The effectiveness of the method is demonstrated in simulations and in processing of measured OCT signals that contain fiel… Show more

“…Light scattering spectroscopy (LSS) was incorporated in OCT for depth resolved nuclear morphology measurements 11,12 . Spectroscopic information was extracted from OCT data 13 , angular resolved low coherence interferometry was demonstrated for the assessment of subcellular features 14 , and molecular imaging was realized 15 . Interferometric synthetic aperture microscopy (ISAM) has been developed to improve the out of focal plane resolution 16 .…”

“…A recently developed frequency transform approach, called the dual-window method, simultaneously utilizes a broad and narrow transformation window in order to obtain high resolution spectroscopy measurements without any tradeoff in depth resolution. 34 The dual window (DW) method was implemented for fLCI and accurately characterized phantoms containing micrometer-sized particles 35 and tissue samples. 36 Figure 7 illustrates the added contrast obtained with spectral analysis using the DW method, comparing a standard OCT image of the microsphere phantom composed of two different sizes with the DW method applied to fLCI (Fig.…”

“…Light scattering spectroscopy has been used in OCT to study the three-dimensional morphology of the cell nuclei [17,18]. Spectral characteristics of the objects were derived from OCT [19], and the authors of [20] studied molecules. A correlation method for imaging the vascular system has been recently developed at our laboratory [21].…”
