Dupuytren's: a systems biology disease

Abstract: Dupuytren's disease (DD) is an ill-defined fibroproliferative disorder of the palm of the hands leading to digital contracture. DD commonly occurs in individuals of northern European extraction. Cellular components and processes associated with DD pathogenesis include altered gene and protein expression of cytokines, growth factors, adhesion molecules, and extracellular matrix components. Histology has shown increased but varying levels of particular types of collagen, myofibroblasts and myoglobin proteins in … Show more

“…This is in part attributable to increased activity in the fibrogenesis research community and in part attributable to the introduction of new minimally invasive treatments. Recent review articles reflect both a greater focus on genetics and 1,2 molecular biology [3][4][5][6] and an openness to alternative treatments. [7][8][9][10] …”

“…Further proteins are platelet-derived growth factor (PDGF), fibroblast growth factor (FGF), epidermal growth factor (EGF) and interleukin-1 (IL-1), as well as the cell matrix proteins Tenascin and Periostin that also affect differentiation, growth and contractility of myofibroblasts. However, the exact pathogenesis and pathology have yet to be decrypted [1,[9][10][11][12].…”
