Dupuytren's disease: Personal factors and occupational exposure

Abstract: Background The etiology of Dupuytren's disease is unknown, and the role of occupational exposure is still debated. Our objective was to study the association between occupational exposures, personal risk factors and Dupuytren's disease. Methods In this cross‐sectional survey, nine occupational physicians performed clinical examinations, focused on Dupuytren's disease, of 2,406 French male civil servants employed at the Equipment Ministry in 1998 and interviewed them about medical history, leisure manual exposu… Show more

“…We know that in the UK, there are a number of different generic codes for DD including the umbrella term musculoskeletal disorders. Six studies [16,26,45,58,73,99] related the epidemiological findings to the heritable nature of the disease. Finsen in 2002 noted that there was a higher prevalence of DD in family members living within the same geographical area as their diseased relative.…”

“…[27][28][29][30]41,42 POSTN upregulation has been associated with various cancers, including melanoma, pancreatic cancer, and oral cancers. 50,51 The upregulation of POSTN, however, may be an effect rather than the cause of DD development. Shimazaki and Kudo demonstrated that POSTN may negatively regulate tumor growth by promoting capsule formation.…”

“…Over-expression of this gene is associated with abnormal cell growth and with various cancers such as melanoma and oral and pancreatic cancers. 8,12,40,41 In addition to abnormal proliferation of fibroblasts, Dupuytren's contracture is characterized by overproduction of collagen and extracellular matrix. Genes related to collagen synthesis (COL13A1, COL5A2), associated with Dupuytren's contracture in several studies, 9,42 are targets of miR-29c.…”
