Dynamic expression of Tbx2 subfamily genes in development of the mouse reproductive system

Abstract: Background Tbx2, Tbx3, Tbx4, and Tbx5, members of the Tbx2 subfamily of T-box transcription factor genes, are important for many aspects of embryonic development and mutations in some human TBX2 subfamily genes cause developmental syndromes. In addition, TBX2 and TBX3 are overexpressed in a variety of cancers, including reproductive system cancers. This study characterizes the expression of Tbx2 subfamily genes during development of the reproductive system. Results We show that these genes are expressed in b… Show more

“…Like many T-box genes, Tbx3 is expressed in a highly dynamic pattern throughout development. Notably, it is present in the developing limbs, mammary gland, craniofacial region, heart, kidney, Wolffian duct epithelium, and mesenchyme around both Wolffian and Müllerian ducts, pituitary gland, dorsal root ganglia, yolk sac, and lungs (Chapman et al, 1996;Douglas et al, 2012). Expression is found in the embryonic precursors of many -if not all -of the tissues affected in UMS, as well as in a number of other tissues not known to be affected in patients with UMS.…”

“…Antisense probes for Tbx2, Tbx4, Tbx5, and Pou5f1 were used as previously described [10], and sections were counterstained with Nuclear Fast Red (Sigma) unless otherwise indicated. ISH with sense RNA controls showed no background staining.…”

“…Tbx4 expression in the bipotential embryonic gonad is first detected at E11.5 [10]. To determine if Tbx4 is expressed in embryonic germ cells or in the surrounding tissue, we analyzed Tbx4 and Pou5f1 expression following busulfan treatment of pregnant females.…”
