In this study hydrogels synthesized from agarose and carbomer 974P macromers were selected for their potential application in spinal cord injury (SCI) repair strategies following their ability to carry cells and drugs. Indeed, in drug delivery applications, one of the most important issues to be addressed concerns hydrogel ability to provide a finely controlled delivery of loaded drugs, together with a coherent degradation kinetic. Nevertheless, solute effects on drug delivery system are often neglected in the large body of literature, focusing only on the characterization of unloaded matrices. For this reason, in this work, hydrogels were loaded with a chromophoric salt able to mimic, in terms of steric hindrance, many steroids commonly used in SCI repair, and its effects were investigated both from a structural and a rheological point of view, considering the pH-sensitivity of the material. Additionally, degradation chemistry was assessed by means of infrared bond response (FT-IR) and mass loss.

Topical corticosteroids are the cornerstone therapy for many inflammatory skin conditions. In recent decades, the development of new molecules with improved benefit/risk ratio and innovative galenic formulations has boosted the field of topical corticosteroid therapy. Alternative classifications of topical corticosteroids have been introduced taking into consideration the potential adverse effects of each molecule in order to provide useful information for clinical practice. Concurrently, new therapeutic regimens with topical corticosteroids have been proposed to provide longterm approaches for the management of chronic relapsing inflammatory skin diseases.The aim of this review is to provide an overview of topical corticosteroids focusing on the recent research performed in the fields of classification methods, galenic formulations and therapeutic strategies.

Topical Oleaginous Minocycline Foam: Efficacious Delivery into Skin Layers

Minocycline hydrochloride (MCH) is a broad spectrum antibiotic of the tetracycline group that is extensively used to treat skin conditions such as acne and rosacea. Unfortunately, since this antibiotic is highly prone to degradation, no minocyclinecontaining topical product is currently available on the market. In this pilot study, MCH was incorporated into a novel oleaginous foam recently developed by Foamix Pharmaceuticals, Ltd. The product, coded FMX101, was produced in drug concentrations of 1% w/w or 4% w/w. An in vitro porcine skin model was utilized to evaluate the extent of MCH penetration into the skin layers and compartments. The results indicated that topical foam administration allows MCH penetration that is sufficiently deep to treat most typical skin infections, acne and rosacea. Additional studies, using MALDI FTICR imaging and fluorescent microscopy techniques demonstrated visually the residence of MCH in the stratum corneum, the epidermis and the dermis and showed specific targeting to the pilosebaceous unit. The antibiotic foam caused virtually no trans-dermal drug absorption.

Abstract: Recent literature suggests that when foams are properly engineered using the advances of in situ analysis techniques, the enhancement of topical drug delivery via engineering this type of vehicle can be achieved.
