Dynamic protein expression of NF‑κB following rat intracerebral hemorrhage and its association with apoptosis

Abstract: The aim of the present study was to evaluate the dynamic protein expression of nuclear factor (NF)-κB and apoptosis in the cerebral tissue surrounding hematoma following intracerebral hemorrhage (ICH) in rats. A total of 80 healthy male Wistar rats were divided into a sham-surgery group and an ICH group. The ICH model was established by injecting autogenous non-heparin anticoagulant arterial blood into the caudate putamen. NF-κB levels were assessed by immunohistochemistry at different time points subsequent t… Show more

“…Secondly, as for all inducible transcription factors, NF-κB stimulation, similar as to Nrf2, may only persist for a limited amount of time, and measurements may be more time-sensitive than the resulting downstream cytokines. This was shown in a rat model of cerebral haemorrhage, where NF-κB protein expression was measured 3 h following injury (cytoplasm), identified in the nucleus at 6 h, with a peak expression of 72 h, lasting for about 5 days [ 447 ]. Finally, expression may differ between cells in the body and measuring NF-κB may require a fair amount of cells.…”

“…7 B1 ). Nf‐κb is a key nuclear transcription factor involved in apoptosis (57). Key signaling components of the NF‐κB pathway, including Tnf‐α , receptor‐interacting protein (Rip)1, RING finger domain protein Traf2/5 , and transcriptional activators P50/P65 , were up‐regulated in VEGF‐deprived retina.…”

“…The NF-jB levels then peaked at 3 days and NF-jB was still present at 5 days. 44 The induction of NF-jB subsequently up-regulated the expression of several genes, including those from the matrix metalloproteinase (MMP) family, NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome and several pro-inflammatory cytokines such as interleukin 1 beta (IL-1b) and tumor necrosis factor-alpha (TNF-⍺). Expression of these genes ultimately leads to neuroinflammation and BBB hyperpermeability.…”
