The maternal immune activation produced by the systemic administration of lipopolysaccharide (LPS) in rats provides valuable insights into the basis of behavioural schizophrenia-like disturbances and biochemical changes in the brains of the offspring, such as microglial activation. Regarding therapy, antipsychotics continually constitute the cornerstone of schizophrenia treatment. To their various efficacy and side effects, as well as not fully recognised mechanisms of action, further characteristics have been suggested, including an anti-inflammatory action via the impact on neuron–microglia axes responsible for inhibition of microglial activation. Therefore, in the present study, we sought to determine whether chronic treatment with chlorpromazine, quetiapine or aripiprazole could influence schizophrenia-like behavioural disturbances at the level of sensorimotor gating in male offspring prenatally exposed to LPS. Simultaneously, we wanted to explore if the chosen antipsychotics display a positive impact on the neuroimmunological parameters in the brains of these adult animals with a special focus on the ligand-receptor axes controlling neuron–microglia communication as well as pro- and anti-inflammatory factors related to the microglial activity. The results of our research revealed the beneficial effect of quetiapine on deficits in sensorimotor gating observed in prenatally LPS-exposed offspring. In terms of axes controlling neuron–microglia communication and markers of microglial reactivity, we observed a subtle impact of quetiapine on hippocampal Cx3cl1 and Cx3cr1 levels, as well as cortical Cd68 expression. Hence, further research is required to fully define and explain the involvement of quetiapine and other antipsychotics in Cx3cl1-Cx3cr1 and/or Cd200-Cd200r axes modulation and inflammatory processes in the LPS-based model of schizophrenia-like disturbances.

Ovа studijа se bavi ispitivanjem potencijаlno zaštitinih efekata аgmаtina (AGM) nа razvoj oksidаtivnog/nitrozativnog stresa u selektivno osetljivim strukturama mozga pacova nakon davanja hlorpromаzina (HPZ). Sve ispitivane supstance aplikovane su u pojedinačnoj dozi intraperitonealno (i.p.). Životinje su podeljene na kontrolnu (K, 0.9 % fiziološki rastvor), HPZ (HPZ, 38.7 mg/kg TM), HPZ+AGM (AGM, 75 mg/kg TM odmah nakon HPZ, 38.7 mg/kg TM i.p.) i AGM (AGM, 75 mg/kg TM) grupu i žrtvovane dekapitacijom 24 h nakon odgovarajućeg tretmana. Rezultаti pokаzuju dа primena HPZ sa AGM značajno smanjuje koncentraciju leka u mozgu pacova u poređenju sа HPZ-životinjаmа (p<0.05). Aplikacija HPZ povećava lipidnu peroksidaciju (p<0.001 u korteksu, strijatumu i hipokampusu), koncentraciju nitrita i nitrata (p<0.001 u sva tri ispitivana regiona mozga) i stvaranje superoksidnog anjon radikala (p<0.05 u sve tri moždane strukture) u odnosu na odgovarajuću kontrolu, dok istovremeno utiče na oštećenje enzimskog antioksidativnog sistema (superoksid dizmutaza u korteksu i strijatumu p<0.05, odnosno hipokampusu p<0.001; glutation reduktaza u kori mozga i strijatumu p<0.001, odnosno hipokampusu p<0.05; katalaza u korteksu p<0.001, odnosno

Guinea pigs (Cavia porcellus) were treated with haloperidol (HP), and free radical (FR) and ferric reducing antioxidant power (FRAP) assays were used to determine oxidative stress levels. Furthermore, the superoxide dismutase (SOD), glutathione reductase (GR) and glutathione-S-transferase (GST) activity levels were detected and glucose levels and the reduced and oxidized glutathione (GSH/GSSG) ratio were measured in HP-treated and untreated guinea pigs. The present study demonstrated that the administration of HP causes significant oxidative stress in guinea pigs (P=0.022). In animals treated with HP, the activity of GST was significantly increased compared with a placebo (P= 0.007). The elevation of SOD and GR activity levels and increase in the levels of glutathione (GSH) in HP-treated animals were not statistically significant. In the HP-untreated animals, a significant positive correlation was observed between oxidative stress detected by the FR method and GST (r=0.88, P=0.008) and SOD (r=0.86, P= 0.01) activity levels, respectively. A significant negative correlation between the levels of plasma glucose and oxidative stress detected by the FRAP method was observed (r=−0.78, P=0.04). Notably, no significant correlations were observed in the treated animals. In the HP-treated group, two subgroups of animals were identified according to their responses to oxidative stress. The group with higher levels of plasma HP had higher enzyme activity and reactive oxygen species production compared with the group with lower plasma levels of HP. The greatest difference in activity (U/μl) between the two groups of animals was for GR.
