EARLY FLOWERING4 Recruitment of EARLY FLOWERING3 in the Nucleus Sustains the Arabidopsis Circadian Clock

Abstract: The plant circadian clock is proposed to be a network of several interconnected feedback loops, and loss of any component leads to changes in oscillator speed. We previously reported that Arabidopsis thaliana EARLY FLOWERING4 (ELF4) is required to sustain this oscillator and that the elf4 mutant is arrhythmic. This phenotype is shared with both elf3 and lux. Here, we show that overexpression of either ELF3 or LUX ARRHYTHMO (LUX) complements the elf4 mutant phenotype. Furthermore, ELF4 causes ELF3 to form foci … Show more

“…In diurnal conditions, PIF4 and PIF5 transcript levels start to rise at midday through the night, with a peak at dawn (Nozue et al, 2007). The evening complex (EC) formed by ELF3, ELF4, and LUX is necessary for this expression pattern Herrero et al, 2012;Lu et al, 2012). LUX has been found to directly bind to PIF4/5 promoters through the LUX binding site (GATA/TCG) and recruit ELF3 and ELF4.…”

“…2G,3A,and 6,A and B). ELF4 forms part of the Evening Complex (Nusinow et al, 2011), which directly represses expression of clock genes such as PRR7, PRR9, GI, and ARRHYTHMO (LUX; Herrero et al, 2012;Mizuno et al, 2014;Box et al, 2015). Intriguingly, ELF4 also is necessary for red light-mediated induction of CCA1 and LHY (Kikis et al, 2005).…”

“…The proteins encoded by these genes form a complex termed the evening complex (EC), in which the ELF3 protein is suggested to act as a molecular scaffold and signaling hub, connecting ELF4 with LUX ). ELF4 appears to have a role in determining the subcellular location of the complex (Herrero et al, 2012) and, thus, possibly in directing the activity of the LUX transcription factor. Loss of any one of these proteins compromises the EC function , an interaction that is likely to explain the generally similar phenotypes of elf3, elf4, and lux mutants.…”
