Effect of 17 β-Estradiol on Phosphoinositide Metabolism and Prolactin Secretion in Anterior Pituitary Cells

Abstract: The present study was undertaken to investigate the effect of 17 β-estradiol (E2) administration on in vitro prolactin (PRL) release and intracellular phosphoinositide metabolism. The incorporation of [3H]inositol (Ins) into phosphatidylinositol (PtdIns), phophatidylinositol-4-phosphate [PtdIns(4)P] and phosphatidylinositol-4,5-bisphophate [PtdIns(4,5)P2], and the generation of inositol phosphate (InsP2) following thyrotropin-releasing hormone (TRH) stimulation were … Show more

“…The relationship between estrogens, in particular estradiol, and prolactin have been well known for some time. The direct actions of estrogens stimulating prolactin release are well documented in the literature [ 43 , 44 , 45 ]. In fact, estradiol is an important regulator of prolactin synthesis [ 46 ].…”

“…The frozen monolayers were scraped free in 0-5 ml KC1 (0-2 mol/l)/EGTA (5 mmol/1), pH 7-4, and total inositol phosphates were extracted in an acidified aqueous-organic solvent system (chloroform/methanol/water/hydrochloric acid, 2:2:1:002 by vol.). They were separated by Chromatographie analysis on Dowex AG 1-X8 anion exchange formate resin as described previously (Judd et al 1987;Kubota et al 1989). In the aqueous phase, free [3H]inositol was initially removed with 01 mol HCOOH/1, and the total labelled inositol phosphates fractions were then eluted with HCOOH (01 mol/l)/NH4 COOH (1-0 mol/1).…”
