Effect of 26 week magnetic field exposures in a DMBA initiation–promotion mammary gland model in Sprague–Dawley rats

Abstract: Several studies have suggested that exposure to 50 Hz magnetic fields promote chemically induced breast cancer in rats. Groups of 100 female Sprague-Dawley rats were initiated with a single 10 mg gavage dose of 7,12-dimethylbenz[a]anthracene (DMBA) at 50 days of age followed by exposure to ambient fields (sham exposed), 50 Hz magnetic fields at either 1 or 5 Gauss (G) field intensity or 60 Hz fields at 1 G for 18.5 h/day, 7 days/week for 26 weeks. A vehicle control group without DMBA was included. Rats were pa… Show more

“…Driven by concerns raised by epidemiologic studies suggesting a potential cancer risk after exposures to ELF-EMF field strengths above the average exposure of 2 mG (0.2 µT) (6,7,30), this study was designed to determine whether there was a biological effect relevant to the carcinogenic process. We used an in vitro cell differentiation system to determine if ELF-EMF could alter chemically induced differentiation and proliferation of Friend erythroleukemia cells.…”

“…Then, as before, these animals were continuously exposed to the MF during mating, gestation, and birth of the third generation. The total numbers of mice in the MF-exposed group evaluated in each generation were (29)(30)(31)(32)(33)(34)(35). The conditions under which these newly available experiments were conducted and the results obtained are in Table 2.…”

“…The 26-week study (33,35) was conducted using a single dose of DMBA (10 mg/rat) prior to MF exposure in contrast to the fractionated dose protocol used previously. This exposure protocol resulted in a tumor incidence (as determined by palpation) at 13 weeks of approximately 65% and at 26 weeks of 100%.…”

“…However, attempts of other groups to replicate our findings failed [19,20,21], which led us to suggest that genetic differences between substrains of SD rats used in our and other studies may be involved [22]. This prompted us to compare different substrains of SD outbred rats in our laboratory in respect to MF effects on cell proliferation in the mammary gland, susceptibility to DMBA-induced mammary cancer, and MF effects on mammary tumor development and growth in the DMBA model [18].…”
