Effect of a Mutant Danbaekkong Allele on Soybean Seed Yield, Protein, and Oil Concentration

Abstract: There is a known negative correlation between soybean [Glycine max [L.] Merr.] seed protein and oil and between protein and yield. This challenges breeders to increase protein concentration while maintaining oil concentration and yield. The objective of this study was to determine if marker-assisted selection for the Danbaekkong (Dan) protein allele on chromosome 20 influences seed yield and quality traits in near isogenic genetic backgrounds. A population of 24 F 7 -derived near isogenic lines (NIL) of soybea… Show more

“…Seeds of the parents and RILs, each with two replications, were planted in a randomized block design at the USDA-ARS farms in Beltsville, Maryland during the years 2012 and 2015. The highly homozygous (>99%) near-isogenic lines (NILs) were generated from a F 7 plant heterozygous for POWR1 from a cross of G03-3101 × LD00-2817P 45 . The NILs homozygous at the POWR1 locus were planted in replications based on randomized complete block design across nine field environments (one in Arkansas, Missouri, North Carolina, and six in Tennessee) during the years 2016 and 2017.…”

“…Approximately 50 seeds per line were analyzed and measured twice. For NILs, ~20 g seeds were grounded to powder and also measured with Perten DA 7250 analyzer 45 . Seed trait measurements were averaged over all replications and locations for both NIL groups and compared.…”

“…This protein allele provides an alternative source of high protein in addition to G. soja. Cunicelli et al (2019) adapted an SNP protocol for protein concentration developed by Warrington et al (2015) to use marker-assisted selection (MAS) for the Dan allele. They concluded that the lines with the Dan protein allele contained significantly more protein, less oil, and lower yield, which supported the results of previous studies regarding the overall negative correlation between protein and oil.…”

“…Warrington et al (2015) reported a significant QTL on Chr 20 in a Benning × Danbaekkong recombinant inbred line (RIL) population, which had an additive effect of 13.6 g kg −1 . Cunicelli et al (2019) created 12 high‐protein lines with the Danbaekkong allele and 12 normal protein lines derived from a single F 7 plant heterozygous at the Chr 20 locus. They reported 31 g kg −1 more protein on average but lower yield for the lines carrying the Danbaekkong allele across nine environments.…”
