Effect of acetone extract of Rumex japonicas Houtt on hydrogen peroxide-induced apoptosis in rat myocardial cells

Abstract: Purpose: To investigate the protective effect of the acetone extract of Rumex japonicas Houtt. (AER) on rat myocardial cells. Methods: R. japonicas was extracted with 75 % aqueous ethanol by reflux to afford total extract (TER H2O2 (p < 0.05, p < 0.01, p < 0.01, respectively). AER (50, 100 and 200 μg/mL) decreased LDH and CK contents of H9c2 cells (p < 0.01). The levels of SOD (p<0.01) and CAT (p < 0.01) were increased by AER treatments (100 and 200 μg/mL); in addition, AER (50, 100 and 200 μg/mL) decreased… Show more

“…The acetone extract of R. japonicus showed protective activity against myocardial apoptosis, through the regulation of oxidative stress levels in cardiomyocytes (LDH, MDA, CK, SOD) and the suppression of the expression of apoptosis proteins (caspase-3, Bax, Bcl-2) on in vitro H 2 O 2 -induced myocardial H9c2 cell apoptosis [175].…”

“…The findings indicated RJH mitigated the trend of outflow of CK and LDH from the cytoplasm by H 2 O 2 ‐induced cell membrane damage; Intriguingly, the levels of superoxide dismutase and catalase were dramatically mounting at the concentration of 100 and 200 μg/mL while malondialdehyde level was dropping after RJH treatment at the concentration of 50, 100, and 200 μg/mL; As apoptotic‐related proteins, after RJH treatment, the expression of caspase‐3 and Bax was downregulated while the Bcl‐2 expression was upregulated. Taken together, acetone extract of RJH reverses the myocardial apoptosis in H 2 O 2 ‐induced H9c2 cells under the probable mechanism of inhibition of oxidative stress and mitochondria‐mediated apoptosis (Zhou et al, ).…”

“…Previous reports have shown that natural extracts/compounds from plants are important resources for finding novel and effective drugs to treat various intractable diseases [16][17][18]. Flavones are reported to possess significant pharmacological activities including anti-sterility, anti-inflammatory, and anti-cancer activities [19,20].…”
