Summary The aim of the study was to investigate the effect of osmolality of the activating solution (AS) on in vitro fertilization of the eggs of common carp, Cyprinus carpio L. Four different AS (NaCl solutions) were used with different osmolalities: AS‐0 (distilled water), AS‐100 (100 mOsm kg−1), AS‐200 (200 mOsm kg−1) and AS‐300 (300 mOsm kg−1). Fertilization was performed in Petri dishes using freshly collected sperm. Insemination (sperm addition to activated eggs) was preceded by exposure of egg samples to each AS for 0 (control group), 15, 30, 60, 90, 120, 150, 180, 210, 240, 270, 300, 330 and 360 s. Embryonic survival was determined 24 h after incubation at 20°C. Additionally, motility parameters (motility rate [MOT, %] and curvilinear velocity [VCL, μm s−1]) of the spermatozoa activated with the tested AS were measured using a CASA (computer‐assisted sperm analysis) system. Motility duration (DUR, s) in each AS was also determined. The results revealed a significant effect of osmolality of the AS on the fertilizing capacity of common carp eggs. There were no viable embryos in the group AS‐300. In the group AS‐0, the eggs lost their ability for fertilization after 90 s; in the remaining groups, eggs could be fertilized for up to 150 s (group AS‐100) or 360 s (group AS‐200). Fertilization failure in the group AS‐300 was caused by the lack of spermatozoa activation in this solution. The present study findings for in vitro common carp fertilization indicate that special attention should be given to the AS osmolality, where 100 mOsm kg−1 was found to be the most suitable.

Within the study, the in vitro fertilization (ivF) in crucian carp, Carassius carassius (L.), with the use of three activating solutions (AS) [reverse osmosis water (RO), Woynarovich (WS) and Billard (BS) solutions] as well as the period of the capacity of eggs to be fertilized (up to 180 s post-egg-activation) in those solutions. Moreover, CASA analysis of sperm motility was conducted with each AS. In control groups (0 s), the highest (P \ 0.05) fertilization rate (93.2 %) was observed with WS. The application of RO and BS affected lower (P \ 0.05) embryo survival (63.5 and \5 %, respectively). High fertilization rate (over 90 %) was recorded up to 30 (RO) and 90 s (WS) post-egg-activation. Progressive sperm motility (pMOT) was high (over 48 %) in all treatment groups up to 30 s. In RO and WS groups, pMOT significantly decreased (P \ 0.05) after 45 s post activation. BS affected pMOT over 10 % up to the 165 s. No differences (P [ 0.05) between the groups were found considering the curvilinear velocity (VCL) 15 s postsperm-activation. Between 30 and 135 s, the lowest (P \ 0.05) VCL after application of RO was noted. The highest VCL (P \ 0.05) up to 135 s was noted after application of BS and WS. All AS activated crucian carp sperm. The lowest fertilization rate was noted when BS was used, despite the high pMOT and VCL. It suggest that the AS which activates the sperm properly may not activate the eggs probably due to high osmolality of BS (262 mOsm kg -1 ). Because eggs retained the longest period of activity in WS, this AS is suggested for ivF in crucian carp.

Summary Compared were the volume and quality of carp Cyprinus carpio L. obtained from semen of males not subjected to hormonal stimulation (N = 10) but injected with 0.9% NaCl in the amount of 1 ml kg−1 b.w., as well as those stimulated using Ovopel (N = 10) in the amount of 1 granule kg−1 b.w. Semen was collected from the males 24 h after injection of hormonal preparation, to determine the pH, total volume (TVS) and volume recalculated per kg of body mass, concentration and total sperm production (TSP). Differences in sperm motility parameters between fish groups were determined by a computer‐assisted sperm analysis (CASA) system. Sperm motility was activated using 10 mm Tris buffer, containing 100 mm NaCl (pH 9.0, osmolality 200 mOsm kg−1). The pH, osmolality, total protein concentration and activity of acidic phosphatase (AcP), lactate dehydrogenase (LDH), ß‐N‐acetylglucosaminidase and arylsulfatase (AS) were determined in the seminal plasma of both test groups. The stimulation of male carp using Ovopel did not significantly affect CASA parameters, but did lead to an over ten‐fold increase in the values of TVS and VOS and nearly six times higher values of TSP as compared to the Control group. A significant increase in pH following stimulation with Ovopel indicates that cyclic AMP increased, thanks to which sperm acquire a capacity to move during their passage through the sperm ducts. An increase in the values of CASA parameters is a likely reflection of these changes.
