Effect of Avène Spring Water on the Activation of Rat Mast Cell by Substance P or Antigen

Abstract: The biological activity of Avène water from two different springs (‘Sainte Odile’ and ‘Val d’Orb’) was studied in vitro on rat peritoneal mast cell activation. A dilution-dependent inhibition of both histamine and prostaglandin D2 antigen-induced release was observed when cells were preincubated with both Avène spring waters. They also inhibited histamine release triggered by substance P. The ability of Avène water to inhibit mast cell activation in vitro may be related with its antiallergic and ant… Show more

“…Dolomiae). It has anti-inflammatory and immunomodulatory properties (Boisnic, Branchet-Gumila, & Segard, 2001;Castex-Rizzi, Charveron, & Merial-Kieny, 2011;Joly et al, 1998;Merial-Kieny, Castex-Rizzi, et al, 2011;Merial-Kieny, Mengual, et al, 2011), which are potentially responsible for its soothing, antipruritic and softening effects on the skin. The anti-inflammatory properties of Avène thermal spring water have been well documented in the treatment of severe skin disorders such as psoriasis, atopic dermatitis or inherited ichthyosis (Bodemer et al, 2011;Merial-Kieny, Castex-Rizzi, et al, 2011;Merial-Kieny, Mengual, et al, 2011), as well as in the healing of facial skin after laser resurfacing (Sulimovic et al, 2002).…”

“…Peritoneal mast cells from tnale Wistar rats were selected as they are easily separated and purified, and as their immune response is similar to that of human skin mast cells [5]. Cell suspensions (2 x lo4/ mL) in Tyrode's buffer were prepared as described in an earlier paper [4].…”

“…Dermatologists agree to assign to ASW a prominent soothing effect towards skin inflammatory or allergic disorders [2]; this led naturally to the undertaking of an experimental study into the influence of this mineral water on the cutaneous immune system. After preliminary experiments showing that ASW restrains human basophil degranulation [3], an 80 ' %I inhibition of both histamine and prostaglandin D, release was observed on rat mast cells incubated with ASW prior to their activation with substance P or with a specific antigen [4].…”
