Radiation vasculitis is one of the most common detrimental effects of radiotherapy for malignant tumors. This is developed at the vasculature of adjacent organs. Animal experiments have showed that transplantation of mesenchymal stem cells (MSCs) restores vascular function after irradiation. But the population of MSCs being engrafted into irradiated vessels is too low in the conventional models to make assessment of therapeutic effect difficult. This is presumably because circulating MSCs are dispersed in adjacent tissues being irradiated simultaneously. Based on the assumption, a rat model, namely, RT (radiation) plus TX (transplantation), was established to promote MSC homing by sequestering irradiated vessels. In this model, a 1.5 cm long segment of rat abdominal aorta was irradiated by 160kV X-ray at a single dose of 35Gy before being procured and grafted to the healthy counterpart. F344 inbred rats served as both donors and recipients to exclude the possibility of immune rejection. A lead shield was used to confine X-ray delivery to a 3 cm×3 cm square-shaped field covering central abdominal region. The abdominal viscera especially small bowel and colon were protected from irradiation by being pushed off the central abdominal cavity. Typical radiation-induced vasculopathy was present on the 90th day after irradiation. The recruitment of intravenously injected MSCs to irradiated aorta was significantly improved by using the RT-plus-TX model as compared to the model with irradiation only. Generally, the RT-plus-Tx model promotes MSC recruitment to irradiated aorta by separating irradiated vascular segment from adjacent tissue. Thus, the model is preferred in the study of MSC-based therapy for radiation vasculitis when the evaluation of MSC homing is demanding.

The tumor microenvironment is essential for the formation and development of tumors. Cytokines in the microenvironment may affect the growth, metastasis and prognosis of tumors, and play different roles in different stages of tumors, of which transforming growth factor β (TGF-β) and tumor necrosis factor α (TNF-α) are critical. The two have synergistic and antagonistic effect on tumor regulation. The inhibition of TGF-β can promote the formation rate of tumor, while TGF-β can promote the malignancy of tumor. TNF-α was initially determined to be a natural immune serum mediator that can induce tumor hemorrhagic necrosis, it has a wide range of biological activities and can be used clinically as a target to immune diseases as well as tumors. However, there are few reports on the interaction between the two in the tumor microenvironment. This paper combs the biological effect of the two in different aspects of different tumors. We summarized the changes and clinical medication rules of the two in different tissue cells, hoping to provide a new idea for the clinical application of the two cytokines.

Although many studies have found that two essential fatty acids, linoleic acid (LA; n‐6 polyunsaturated fatty acid, PUFA) and α‐linolenic acid (ALA; n‐3 PUFA), are closely related to the occurrence and development of cardiovascular disease (CVD), the effects of LA and ALA ratios and concentrations on CVD risk are not well known. The aim of this study is to investigate the effects of various LA/ALA ratios (1:1, 5:1, 10:1, and 20:1) and their concentrations on inflammation, oxidative stress, endothelial cell functions, and the mechanism in human umbilical vein endothelial cells (EA.hy926 cell line). The results indicate that low LA/ALA ratio might provide CVD benefits at low PUFA concentration while low LA/ALA ratio (1:1) increases CVD risk at high PUFA concentration. The results show that the effects of LA and ALA on inflammation, oxidative stress, endothelial cell functions, and the related gene expression are influenced by not only LA/ALA ratios but also their concentrations, and the mechanism may be mediated through PKCα/NF‐κB inflammatory and NO/eNOS signaling pathways by regulation of the NOX4 pathway. Practical Applications: Lipid metabolism, inflammation, oxidative stress, and endothelial cell functions play an important role in the pathogenesis of CVD, which may be affected by an imbalance in the LA/ALA ratio. However, the effects of LA/ALA ratios on CVD risk are influenced by PUFA concentrations. The study provides a method to design diets beneficial for CVD prevention. The low LA/ALA ratio (1:1) could up‐regulate increase the gene expression level of PKCα and eNOS gene while NF‐κB and NOX4 gene expression levels are up‐regulated to increase oxidative stress, which could cause the oxidation of BH4 and uncoupling of eNOS under high PUFA concentration. Uncoupling eNOS generated normalO2−, instead of NO, which could damage vascular endothelial cell function. This is different from the role of eNOS in the production of NO under normal physiological conditions, which shows the duality of eNOS on atherosclerosis. The results indicate both LA and ALA ratios and concentrations are important for CVD risk.
