Effect of cisplatin on the expression of glutathione-S-transferase in the cochlea of the rat

Abstract: Glutathione-S-transferase (GST) has been found to conjugate glutathione (GSH) to diverse electrophiles and play a major role in the detoxification of alkylating and platinating agents. However, there is little information regarding the pattern of GST expression in the cochlea. Although cisplatin is ototoxic, its effect on GST in the cochlea is unknown. In the present study we investigated the pattern of GST expression in control and cisplatin-treated cochleas by using the laboratory rat as animal model. Sixtee… Show more

“…GSH system is a key component of the overall defense system that has the capacity of detoxifying both endogenous and exogenous toxic substances and also in regenerating vitamin C and E to their active forms [ 74 , 75 ]. GST is an enzyme involved primarily in the detoxification of highly reactive electrophiles such as drugs by combining with GSH as conjugating agent [ 76 ]. The decrease observed in the levels of AA and GSH as well as activity of GST occasioned by CP may predispose the liver to oxidative stress and tissue injury.…”

“…This is because GSH and AA have the capability to scavenge free radicals and act as the first line of defence against cellular oxidation [ 56 , 57 ]. GST is an enzymatic antioxidant that detoxifies most highly reactive electrophiles, carcinogens and products of oxidative stress in conjugation with GSH [ 58 ]. The importance of the GSH molecule has been linked to its capacity to regenerate active forms of both vitamins C and E, and with the detoxification of endogenous and exogenous toxic substances in the body [ 59 , 60 ].…”

“…Treatment of patients with CP caused a decrease in a highly reactive electrophiles and glutathione, which may be due to the electrophilic burden on the cells and the formation of acrolein, which leads to a decrease in glutathione content [56]. Treatment of rats with nZnO increased glutathione levels in the lung glutathione catalyzes the conjugation of glutathione with a highly reactive electrophiles, which plays a major role in the detoxi ication of alkylating agents [57].…”
