Effect of excision repair system on antibacterial and mutagenic activity of daunomycin and other intercalating agents inSalmonella typhimurium

Abstract: Daunomycin and adriamycin, are more mutagenic and antibacterial for a strain of Salmonella typhimurium defective for the uvrB gene than for its uvr+ counterpart. Other intercalating agents, as some acridine dyes, affect equally the two bacterial strains.

“…Moreover, drug amounts too low to produce any halo on WP2 or TM930 gave quite discernible zones of inhibition on the other strains. Similar results (not shown) were obtained with other mutagens and carcinogens, such as 4-nitroquinoline-1-oxide, hycanthone, ethidium bromide, 1-(5-nitrofurfurylidene)amino-hydantoin (8), K2Cr2O7 (17), cis-platinum dichlorodiamine (11), and daunomycin (8,14). It is worth noting at this point that the antibacterial activity of a drug not mediated by DNA interaction generated equal inhibition zones in all strains including WP2 wild type.…”

“…The genotoxic potentials of these compounds are well documented in the literature. Both ADM and DNM are mutagenic in prokaryotic Pani et al, 1975;Benedict et al, 1977;Seino et al, 1978;Ferguson and Denny, 19791 these studies. The kind gifts of 5-iminodaunomycin by Dr. R. Wolgemuth, Adria Laboratories, and N-acetyldaunomycin by Dr. E. Herman, FDA, are acknowledged.…”

The carcinogenic potentials of ADM and DNM are underscored by the finding that, in rats, a single dose of either compound can result in a tumor incidence of greater than 80% within 1 yr [Bertazolli et al, 1971;Marquardt et al, 1976;Solcia et al, 19781.…”
