Effect of Gum Arabic on Oxidative Stress and Inflammation in Adenine–Induced Chronic Renal Failure in Rats

Abstract: Inflammation and oxidative stress are known to be involved in the pathogenesis of chronic kidney disease in humans, and in chronic renal failure (CRF) in rats. The aim of this work was to study the role of inflammation and oxidative stress in adenine-induced CRF and the effect thereon of the purported nephroprotective agent gum arabic (GA). Rats were divided into four groups and treated for 4 weeks as follows: control, adenine in feed (0.75%, w/w), GA in drinking water (15%, w/v) and adenine+GA, as before. Uri… Show more

“…The changes in the renal structure and in the vascular function in rats with adenine - induced CKD have recently been investigated, [8,9,14,15] but as far as we are aware, there are no reports on similar morphometric analysis of the myocardium in this animal model.…”

“…The proposed mechanism by which GA may have produced a salutary action on the kidney has been discussed earlier [8,17,18,19]. Briefly, GA exerts both anti-inflammatory and anti-oxidant actions, and these may counteract the generation of free radicals and the inflammation that - is associated with adenine -induced CKD.…”

“…A 4 weeks treatment with adenine increased the pro-inflammatory cytokine TNF-α, and, according to glutathione and superoxide dismutase, triggered oxidative stress [40]. Adenine treatment decreased the creatinine clearance and increased the plasma urea and creatinine cocnentrations, effects significantly blunted by additional GA treatment [41].…”

“…Adenine caused marked renal damage, effects ameliorated by GA treatment for four consecutive weeks. GA reversed the decrease in body weight and the glomerular, tubular and interstitial renal lesions following adenine treatment [40]. It has been argued that those effects of GA may favourably influence the clinical course of chronic renal disease [40].…”
