Effect of Bupleuri Radix Extracts on the Toxicity of 5‐Fluorouracil in HepG2 Hepatoma Cells and Normal Human Lymphocytes

Abstract: Despite the excellent chemotherapeutic effect of 5-fluorouracil, its cytotoxicity and genotoxicity in normal cells remain a major problem.

“…In hepatoma cells, 5-FU induced the up-regulation of Bax, p53, and p21 and triggered the collapse of the mitochondria membrane potential [28]. 5-FU also induced expression of the pro-apoptotic proteins (caspases-8, -9, -3, and Bax) in HeLa cervical carcinoma [29].…”

“…Jurkat T-cells were exposed to PEI or PAMAM polymers for 6 hours and then incubated with DiOC 6 (40 nM) for 30 minutes at 37°C. Cells were washed twice with PBS and analyzed by using flow cytometry (FACSCalibur; Becton-Dickinson) (Kang et al, 2008). All experiments were performed at least three times.…”

“…Chaihu, Danggui, and Shanzhi are the three identical herbs used in both TCHYS and LDSGT. Extracts from Chaihu (Bupleuri chinese) may be effective as a therapeutic agent to treat hepatomas only, because it increased mitochondria membrane potential collapse in HepG2 cells, but not in lymphocytes 18 . Danggui (Angelicae sinensis) has been reported to possess immunomodulatory functions, as the treatment of Danggui-containing TCM to cancer patients, who were receiving chemotherapy and/or radiotherapy, showed improvement in their immune functions 5 .…”
