Effect of iron complexes on adjuvant arthritis in rats.

Abstract: When a total dose infusion of iron dextran is given to anaemic rheumatoid patients an exacerbation of inflammatory synovitis in previously affected joints is observed. The adjuvant arthritis model of inflammation in rats has been used to investigate the mechanism of iron promoted synovitis. Either iron dextran (5 mg injected intravenously) with a dextran C control, or iron sorbitol (7.5 mg injected intramuscularly) with a sorbitol citrate complex control was given at the onset of clinical joint inflammation. I… Show more

“…During the 28 days, inflammation was measured subjectively as the Total Arthritic Index, which is a score based on redness, swelling, and deformity of the joint and is similar to the scoring system used in the clinic (28). The results from such an experiment are shown in Fig.…”

“…Changes in rear paw volume were quantified daily by plethysmography using a Buxco Edema Table (Buxco Electronics, Sharon, CT). Joints also were scored daily for symptoms of arthritis by using a standard scoring system modified from Dabbagh et al (28). Each joint could receive a maximum score of 4 or a maximum score of 16 for each animal.…”

“…Eating meat and seafood rich in iron and purine is more likely to cause GA than vegetables rich in purine 9 . Iron supplementation will aggravate synovial inflammation in the arthritis rat model 10 . Iron overload not only inhibits osteoblast differentiation but also promotes the formation and activity of osteoclasts, 11 which suggests that iron overload may play an important role in the pathogenesis of AGA.…”

Therefore, research on iron overload in the pathogenesis of AGA mice was carried out by our team.…”

“…16] as did the k opioid agonist U50488 [17], In adjuvant arthritis 2-alkoxyl-2-aryl-4-alkylmorpholine, a compound with opioid and anti-oxidant properties, was found to be high ly effective at reducing both primary and sec ondary lesions [18]. Both models have been shown to possess a significant ROS compo nent [19,20]. There is evidence, therefore, that opioid receptor agonists can protect against ROS-induced inflammation and our aim in this study is to examine this more directly.…”
