Effect of Juice and Extracts from Saposhnikovia divaricata Root on the Colon Cancer Cells Caco-2

Abstract: Colorectal cancer ranks 3rd in terms of cancer incidence. Growth and development of colon cancer cells may be affected by juice and extracts from Saposhnikovia divaricata root. The objective of the research was to analyze the effect of S. divaricata juice and extracts on the viability, membrane integrity and types of cell death of Caco-2 cells. Juice and extracts were analyzed using Ultra-High Performance Liquid Chromatography-Mass Spectrometry (UHPLC-MS) and in respect of the presence of antioxidants, total c… Show more

“…The human intestinal Caco-2 cell line model was extensively used over past few years to screen bioavailability of elements ions from various food products and beverages 10 , 34 . Here, the cytotoxic effect of NTAP-treated AJ on human colon adenocarcinoma cells (Caco-2) and normal human intestinal epithelial cells (FHs 74 Int) were studied by using the MTT assay in order to reveal any cytotoxic properties of NTAP-treated AJ.…”

“…The levels of glucose (Glu), total protein (TP), albumin (Alb), total cholesterol (TC), and triglyceride (TG) were determined. Glutathione (GSH) was measured quantitively using Ellman’s method modified by Matusiewicz et al [ 31 ]; in this method, 5,5ʹ-dithiobis(2-nitrobenzoic acid) (DTNB, Ellman’s reagent) is reduced by thiol compounds to form a colored product (2-nitro-5-mercaptobenzoic acid) with the maximum absorbance at 412 nm. To 375 µL of serum samples from each group, 19.7 µL of 50% trichloroacetic acid (TCA) was added and centrifuged (1200× g , 5 min).…”

“…The anti-oxidant in vitro properties of extracts were measured using DPPH free radical scavenging test, as previously shown by Matusiewicz et al (2009) . Concentrations ranging from 100 to 1,000 µg/mL were used and the results were expressed as a percentage of DPPH inhibition.…”
