Effect of Milk Renewal on Cell Viability In Vitro at Different Time Frames

Abstract: The purpose of this study was to evaluate if the renewal of milk as a storage medium, every 12, 24 and 48 h, is able to increase its ability to maintain human periodontal ligament fibroblasts (PDLF) viability over time. PDLF were soaked in Minimum Essential Medium at 37 °C (MEM-37) (positive control), tap water (Water) (negative control) and in skimmed milk (44 wells) at 5 °C and 20 °C. The skimmed milk was renewed every 12 h (Milk-12), 24 h (Milk-24) and 48 h (Milk-48) in 11 wells of each plate, and the milk … Show more

“…For the cells stored in milk, their viability levels at 30 minutes and 1 hour were also similar, but there was decline in the cell viability between 1 and 3 hours (p<0.05). The latter observation may be due to a decline in the pH of milk over time 31 . However, the cells stored in egg white had a sequential decline in their viability, similar to the findings of Badakhsh et al 20 (2014), and this accounted for a sequential decline in the nutrients in egg white over time.…”

“…Este produto, possui propriedades fisiológicas, incluindo o pH e a osmolaridade, compatíveis com as células do ligamento periodontal. Além disso, é de fácil disponibilidade, e são livres de bactérias 16,17 . Embora 75% dos professores e 86,11% dos alunos soubessem que tempo e meio de armazenagem são fatores cruciais em casos de avulsão, os resultados mostraram que poucos professores tinham conhecimento sobre o tempo crítico para o reimplante e meio de armazenagem ideal.…”

“…Then DMSO (Dimethylsulfoxide) (100 mL per well) was used to dissolve the precipitated formazan. Finally, the absorbance was measured by a micro-plate reader (Hercules, CA, USA) at 450 nm and the cell viability was determined using the following equation [51]:…”
