Effect of Pre-treatment Solution on Iris hollandica Flower: Vase life, Anthocyanin Content, and Peroxidase Activity

Abstract: Iris hollandica `Blue Magic' was treated with deionazed water as a control, 3% sucrose (Suc), 3% sucrose plus 0.4 mm silver thiosulphate (Suc+STS), 3% sucrose plus 200 mg·L-1 8-hydroxyquinoline sulphate (Suc+HQS) and 3% sucrose plus 100 mg·L-1 benzyl amino-purine (Suc+BA) for 4hrs and then transferred to tap water. “…Similar results were observed in pepper (Capsicum annuum L.) where embryogenesis was improved and one to three plants per 100 anthers were obtained when 4°C temperature for 48 h was applied as a pre-treatment to flower buds (Thriveni et al, 2020). However, a different kind of observation was also documented in pepper and there were no significant effects of cold pre-treatment on flower buds (Vagera & Havranek, 1985;Kim et al, 2004;Jha et al, 2021) The results suggested that various genotypes had different requirement of nutrients and growth regulators. The highest callus induction frequency for anthers from different size of fresh flower buds for different genotypes were TA8(MS + 2iP (0.5 mg L -1 ) + NAA (0.5 mg L -1 )) for LE 1230 and PKM1, TA1(MS + 2iP (1.0 mg L -1 ) + IAA (2.0 mg L -1 )) for CO 3, LE 1238 and LE 1256 and TA7 (MS + 2iP (0.5 mg L -1 ) + Kinetin (1.5 mg L -1 ) + NAA (1.0 mg L -1 )) for TLCV2 and the least was always recorded in TA0 (MS basal).…”
