Effect of prolonged administration of the serotonin4 (5-HT4) receptor agonist cisapride on aldosterone secretion in healthy volunteers

Abstract: In man, serotonin (5-HT) has been shown to stimulate aldosterone secretion through activation of 5-HT4 receptors, In particular, we have observed that oral administration of a single dose of the 5-HT4 receptor agonist cisapride (10 mg) induces a 5-fold increase in plasma aldosterone levels in healthy volunteers. Surprisingly, the usual disorders associated with hyperaldosteronism, i.e. hypertension and hypokalemia, have never been reported during chronic treatment with cisapride. In the present study, we have … Show more

“…5-HT4 receptors are present in the normal adrenal cortex (26) and in the adrenals of patients with AIMAH (20). Accordingly, a strong stimulating effect of serotonergic substances have been described for aldosterone secretion and to a weaker degree for cortisol secretion (31,32).…”

“…Na espécie humana, a serotonina (5-HT) e os agonistas serotoninérgicos, através de mecanismo parácrino, são potentes estimuladores da secreção de aldosterona e em menor intensidade de cortisol (51). Este efeito está relacionado à expressão do receptor da serotonina (5-H T 4 R), principalmente na zona glomerulosa e fracamente na zona fasciculada (52).…”
