Effect of Recombinant Bovine Somatotropin (rbST) on Cytoplasmic Maturation of Bovine Oocytes and Their Developmental Competence In Vitro

Abstract: Abstract. The objectives of this study were to evaluate the effects of recombinant bovine somatotropin (rbST) on the nuclear and cytoplasmic maturation of bovine oocytes and their further developmental competence to blastocysts in vitro. We analyzed the mitochondrial activity and concentration of intracellular stored calcium ([Ca 2+ ]is) in matured oocytes and the morphology and chromatin status of produced embryos after in vitro fertilization. Cumulus-oocyte complexes were incubated in TCM 199 containing 10… Show more

“…Only the labelled mitochondria that were actively respiring were recorded. The fluorescence intensity per oocyte (mA) was measured by the Nikon Photometry System P 100 (Nikon, Düsseldorf, Germany) as described in pig, horse and bovine oocytes (Torner et al 2004, 2007, Kuzmina et al 2007. For measurement of intensity, we placed the whole oocyte (thickness around 20 mm) with the eyepiece of the Photometer head P 100 in a defined area of measurement (same size for all observation).…”

“…It is known that oocytes require maturation of both cytoplasm and nucleus to be able to support successful fertilization and subsequent embryo development (Kuzmina et al, 2007;Trounson et al, 2001).…”
