Effect of Schisandrin B on Hepatic Glutathione Antioxidant System in Mice: Protection against Carbon Tetrachloride Toxicity

Abstract: Pretreating female Balb/c mice with schisandrin B (Sch B) at increasing daily doses (1-4 mmol/kg) for 3 days caused dose-dependent increases in hepatic glutathione S-transferase (GST) and glutathione reductase (GRD) activities. However, the activities of glucose-6-phosphate dehydrogenase (G6PDH), Se-glutathione peroxidase (GPX), and gamma-glutamylcysteine synthetase (GCS) were down-regulated to varying degrees in a dose-dependent manner. While there were biphasic changes in hepatic reduced glutathione (GSH) le… Show more

“…This derivative has been used for the treatment of viral and chemical hepatitis. additionally, several studies have shown that Sch B protects against free radical-mediated hepatocellular damage and enhances hepatic glutathione antioxidant status in the mitochondria (38,39). it has been reported that schisandrin B has enantiomers [(+)Sch B and (-)Sch B] (40), which exhibit differential functions.…”

“…The degree of GSH depletion induced by t-BHP challenge was estimated by comparing the GSH level at 1 min post-t-BHP challenge with the basal value (i.e., at 0 min) and expressed as % depletion. GR and G6DPH activities were measured as described by Seelig and Meister [19] and Ip et al, [20] respectively.…”

“…As a traditional medicinal herb, S. chinensis has been used as an astringent curing dry cough, asthma, night sweats, nocturnal seminal emissions and chronic diarrhea [1,2]. Modern medical research had proved that S. chinensis contains multiple active components used to protect liver [3][4][5], prevent senility [6][7][8][9], restrain oxidation [10,11], improve human body immunity ability [12], regulate the central nervous system and so on [13,14].…”
