Pregnant Wistar rats were daily injected with 5 mg sulpiride into the peritoneal cavity starting on day 14 of gestation. Blood samples were obtained from the maternal carotid artery and fetal axillary vessels on day 20 of gestation. Serum prolactin (PRL) levels were determined by radioimmunoassay using a rat PRL RIA kit. The maternal serum and fetal plasma PRL levels were significantly higher in the sulpiride-treated group than in the saline control group. There was a significant positive correlation between fetal body weight and fetal plasma PRL level. These results suggest that sulpiride which reached the fetus stimulates fetal PRL secretion and that PRL may exert a growth-promoting effect on the fetus.----prolactin; sulpiride; fetal growth; ratThe roles of prolactin (PRL) during pregnancy still remain unclear, especially the effect on the fetus. The PRL levels in fetal circulation increase significantly after 28 gestational weeks in human (Winter et al. 1975), and fetal growth is promoted markedly after the end of mid gestation. Although anencephalic infants have a low growth hormone level, the growth retardation has not been found in them (Furuhashi et al. 1980). These findings suggest that PRL may have some growth promoting effect on human fetus, and that growth hormone is not essential for fetal growth.In this study, we investigated the effect of PRL on the fetal growth and the relationship of PRL and fetal body weight using sulpiride-treated pregnant rats. MATERIALS AND METHODSSulpiride (Dogmatyl®) 5 mg or saline (control) was daily injected into the peritcneal cavity of pregnant Wistar rats from 14 days of gestational age. On day 20 of gestation all animals were sacrificed under ether anesthesia and maternal blood was obtained from the carotid artery, and the then all fetuses were exteriorized. Fetal body weight was measured immediately after exteriorization. Fetal blood samples were collected via an incision from axillary vessels using heparinized capillaries. The blood was pooled from fetuses which were taken out from one side of the uterus.

Poor production of breast milk is the most frequent cause of breast lactation failure. Often, physician prescribe medications or other substances to solve this problem. The use of galactogogues should be limited to those situations in which reduced milk production from treatable causes has been excluded. One of the most frequent indication for the use of galactogogues is the diminution of milk production in mothers using indirect lactation, particularly in the case of preterm birth. The objective of this review is to analyze to the literature relating to the principal drugs used as galactogogues (metoclopramide, domperidone, chlorpromazine, sulpiride, oxytocin, growth hormone, thyrotrophin releasing hormone, medroxyprogesterone). Have been also analyzed galactogogues based on herbs and other natural substances (fenugreek, galega and milk thistle). We have evaluated their mechanism of action, transfer to maternal milk, effectiveness and potential side effects for mother and infant, suggested doses for galactogogic effect, and recommendation for breastfeeding.

RÉSUMÉ. Afin d'établir le rôle joué par la prolactine dans le retentissement de la lactation sur l'évolution de Strongyloides ratti chez la ratte, les auteurs essaient de reproduire les effets de cet état physiologique par un traitement avec le sulpiride (benzamide substitué) dont l'effet prolactinogène est intense chez la ratte. Ils notent que les doses faibles reproduisent partiellement les effets de lactation sur le parasitisme en diminuant son intensité et en retardant le début du dépara sitage sans toutefois empêcher celui-ci. L'action pharmacologique du sulpiride, qui agit en bloquant les récepteurs à la dopamine, permet d'expliquer que seules les doses faibles agissent et qu'elles limitent leur action au début du parasitisme. Cette expérimentation apporte des arguments en faveur de l'hypothèse attribuant à la prolactine un rôle fondamental dans les modifications de l'évolution de Strongyloides ratti pendant la lactation. In order to establish the role played by prolactin in lactation during the evolution of Strongyloides ratti in the female rat, the authors tried to reproduce the effects of this physiolo gical state by treatment with sulpiride (substituted benzamide), of which the prolactinogenic effect is intense in the female rat. Low doses partially reproduced the effects of lactation on parasi tism by diminishing its intensity and delaying the onset of rejection, although without preventing it. The pharmacological action of sulpiride, which acts by blocking the dopamine receptors, explains why only low doses are active and why their action is limited to the onset of parasitism. This experiment lends weight to the hypothesis that prolactin plays a fundamental role in modifi cations of the evolution of Strongyloides ratti during lactation.
