Effect of Yikun Neiyi Wan on the Expression of Aromatase P450, COX-2, and ER Related Receptor in Endometrial Cells in Vitro from Patients with Endometriosis

Abstract: The mechanism by which YKNYW alleviates endometriosis pain is possibly related to the decrease in ectopic endometrial P450 arom and COX-2 expression in glandular epithelium, contrary to gestrinone, and the increase in ER expression in mesenchymalis, consistent with gestrione in patients with endometriosis.

“…Endometriosis is a nonmalignant, chronic and common gynecological disease defined as the implantation of endometrium in an abnormal or ectopic location outside the uterus. [13] The exact prevalence of endometriosis is unclear. [4] It is estimated that it accounts for about 10% to 15% of women in reproductive age around the world, [5] up to 50% of women with infertility.…”

“…This results in higher rates of amenorrhea and pain-free intervals after just one month of treatment. Gestrinone acts in the endometrium by stimulating the androgen receptors, and the ensuing inhibition of aromatase expression may be a consequence of the reduction in inflammation, since gestrinone has been shown to have no direct effect on aromatase expression in cultured cells from endometriotic cysts [16]. Inflammation plays a pivotal role in the progression of endometriosis [11] and activation of the androgen receptors may represent an alternative for those patients in whom progestin therapy is unsuccessful.…”

“…22Y24 The activity of the enzyme varies considerably between men and women 25 with age, 25,26 menstrual cycle, 27 and weight. 28 In addition, its activity may be influenced by many extrinsic factors, including herbal supplements 29 and calcitriol. 30 Because aromatase is present in many tissues, including adipose tissue, breast, bone, brain, liver, and blood vessels, 22 it is difficult, if not impossible, to assess the overall contribution of this enzyme to systemic drug clearance using in vitro studies conducted in any individual tissue.…”
