Summary:The aim of this present study was to investigate the effects of extenders containing different antioxidants upon the principle sperm characteristics of Saanen buck semen following freezing. In this study, four bucks were used. Semen was collected twice a week by an artificial vagina during the breeding season to obtain ten ejaculates per animal. The samples were then extended in three extenders (tris, T-, skimmed milk, M-and laiciphos 488, L-based) without (as controls) or with the two antioxidants (5 mM cysteine, C and 1000 µg/ml hyaluronic acid, H). By this way, nine experimental groups were assigned, as follows:For the T extender groups, the rates of motility and viability were significantly (p<0.01) higher for the T-C extender as compared to those of the T-H and T-O. However, there was no such difference (p>0.05) among the extenders based on the rates of abnormal spermatozoa, abnormal acrosome and hypoosmotic swelling (HOS) test. For the M groups, the motility was significantly (p<0.01) higher for the M-C and M-O extenders as compared to that of the M-H. However, there was no such difference (p>0.05) among the extenders based on the rates of viability, abnormal spermatozoa, abnormal acrosome and HOS test. For the L groups, the rates of motility and viability were significantly (p<0.01) higher for the L-C extender as compared to that of the L-H but not that of the L-O. Additionally, there was no such difference (p>0.05) among the extenders mentioned above based on the rates of abnormal spermatozoa, abnormal acrosome and HOS test. As a result, the present in vitro findings of semen quality parameters studied suggest that; i) both skimmed milk and laiciphos extenders were more favourable than tris, and that ii) the cysteine, as an antioxidant, provided a superior protection than the hyaluronic acid did.

Objectives: As very few studies were done on the freezability of pure Egyptian cattle bull sperm. So, we designed this study to evaluate the individual variations in freezability of native bull semen extended in Tris based diluent and sodium citrate-based diluent. Methods:Semen was collected by artificial vagina, examined at once in farm laboratory. Only semen samples fit the minimum parameters are extended in two extenders first the universal one (TRIS) and second modified Sodium citrate extender by adding glycerol (CU-16) which created at Cornell University to evaluate the individual bull variation and interaction between extender and bull.Results: Post-Thawing individual sperm motility, live, abnormal, acrosome integrity percentage was evaluated in addition to Hypo-Osmotic Swelling test (HOS). The highest value for motility, live abnormal and acrosome percentage were 44.00 ± 1.12, 52.25 ± 1.60, 21.25 ± 0.81 and 62.80 ± 2.58 from bull 2, 1, 3 and 2, respectively for semen extended in TRIS, and 42.25 ± 1.61, 52.00 ± 1.76, 22.15 ± 0.85 and 57.40 ± 3.07, from bull 1, 1, 4, 3, respectively for semen extended in CU-16. The results of HOS were 59.25 ± 1.76 and 55.95 ± 2.13 from bull 1 extended in TRIS and CU-16, respectively. Conclusion:A significant variation (p<0.05) in tested parameter was clear when semen extended in TRIS extender with the absence of such variation when semen extended in CU-16 except in live sperm percentage. With ignoring the extender effect a clear significant variations were detected between bulls in all tested parameters.

The aim of this study was to research the effects of four antioxidants that had been added to semen diluents, on spermatological parameters, anti-oxidant enzymes activities and DNA integrity after the freeze-thaw procedures in Brown-Swiss bull semen. A total of 24 ejaculates were collected from three bulls. Each ejaculate was divided into five equal parts and they diluted to the Tris + 25 mM trehalose base extender containing fetuin 1 mg/ml, dithioerithritol 1 mM, cysteamine 4 mM or linoleic acid 0.5 ml/50 ml, and no additives (control). Group dithioerithritol and linoleic acid showed the higher rates of CASA progressive motility, CASA sperm motility (P<0.05) compared with the other groups. Group linoleic acid provided the better protective effect for acrosome and total abnormalities (P<0.01, P<0.05; respectively). Also, group dithioerithritol showed the highest values for the HOS test (P<0.01). In the comet test, group dithioerithritol enhanced tail with lesser chromatin damage than the other groups (P<0.05). Supplementation of dithioerithritol significantly affected the GSH activity (P<0.01). Also groups dithioerithritol and linoleic acid gave higher CAT values than the other groups (P<0.05). Additionally, supplementation of fetuin and cysteamine showed the lowest total antioxidant activity value (P<0.05). In conclusion, we may said that, the addition of antioxidants, specially dithioerithritol and linoleic acid, to added with 25 mM trehalose Tris extender improve post-thaw sperm parameters. Üç boğadan toplam yirmidört ejakülat toplandı. Her bir ejakülat beş eşit kısma ayrıldı ve 25 mM trehaloz katılmış Tris temel sulandırıcısı içerisinde, fetuin 1 mg/ml, dithioerithritol 1 mM, sisteamin 4 mM veya linoleik asit 0.5 ml/50 ml, ve antioksidan içermeyen (kontrol) ile sulandırıldı. Grup dithioerithritol ve L diğer gruplarla kıyaslandığında daha yüksek CASA progressif motilite ve CASA sperma motilitesi gösterdi (P<0.05). Grup linoleik asit akrozom ve toplam abnormalitede daha iyi koruyucu etki sağladı (sırasıyla P<0.01, P<0.05). Keza, grup dithioerithritol, HOS test için en yüksek değeri verdi (P<0.01).
