Background Decomposition of plant litter is a key driver of carbon and nutrient cycling in terrestrial ecosystems. Mixing litters of different plant species may alter the decomposition rate, but its effect on the microbial decomposer community in plant litter is not fully understood. Here, we tested the effects of mixing with maize (Zea mays L.) and soybean [Glycine max (Linn.) Merr.] stalk litters on the decomposition and microbial decomposer communities of common bean (Phaseolus vulgaris L.) root litter at the early decomposition stage in a litterbag experiment. Results Mixing with maize stalk litter, soybean stalk litter, and both of these litters increased the decomposition rate of common bean root litter at 56 day but not 14 day after incubation. Litter mixing also increased the decomposition rate of the whole liter mixture at 56 day after incubation. Amplicon sequencing found that litter mixing altered the composition of bacterial (at 56 day after incubation) and fungal communities (at both 14 and 56 day after incubation) in common bean root litter. Litter mixing increased the abundance and alpha diversity of fungal communities in common bean root litter at 56 day after incubation. Particularly, litter mixing stimulated certain microbial taxa, such as Fusarium, Aspergillus and Stachybotrys spp. In addition, a pot experiment with adding litters in the soil showed that litter mixing promoted growth of common bean seedlings and increased soil nitrogen and phosphorus contents. Conclusions This study showed that litter mixing can promote the decomposition rate and cause shifts in microbial decomposer communities, which may positively affect crop growth.

The glucosinolates of Brassicaceae plants are converted into bioactive isothiocyanates and other volatiles during a challenge by pathogens and other biotic stressors. However, the role of alternative downstream products with weaker potency (e.g., nitriles) is far from being fully understood. This study tested the possible synergistic antifungal interaction between various glucosinolate-derived nitriles and 2-phenylethyl isothiocyanate (PEITC) on 45 fungal strains, including endophytes from horseradish roots (Brassicaceae) and soil fungi, using an airtight system enabling the accurate study of extremely volatile antifungal agents. The median minimal inhibitory concentrations (MICs) were 1.28, 6.10, 27.00 and 49.72 mM for 1H-indole-3-acetonitrile (IAN), 3-phenylpropanenitrile (PPN), 4-(methylsulfanyl)-butanenitrile (MSBN) and 3-butenenitrile (BN, = allyl cyanide), respectively. Thus, nitriles were considerably weaker antifungal agents compared to PEITC with a median MIC of 0.04 mM. For the same nitriles, the median fractional inhibitory concentration indices (FICIs) of the combinations were 0.562, 0.531, 0.562 and 0.625, respectively. Altogether, 47.7%, 56.8%, 50.0% and 27.3% of tested fungal strains showed a synergistic antifungal activity (FICI ≤ 0.5) for the nitrile–isothiocyanate combinations, respectively. Hypocreales strains showed the least sensitivity towards the GSL decomposition products and their combinations. The mean MIC values for PEITC showed 0.0679 ± 0.0358, 0.0400 ± 0.0214, 0.0319 ± 0.0087 and 0.0178 ± 0.0171 mM for Hypocreales, Eurotiales, Glomerellales and Pleosporales, respectively. In addition, nitriles, especially IAN, also showed significant differences. For the same fungi, the median FICI values fell in the ranges of 0.61–0.67, 0.52–0.61, 0.40–0.50 and 0.48–0.67, respectively, depending on the nitrile. Our results suggest that glucosinolate-derived nitriles may enhance isothiocyanate antifungal activity and that they may play an active role in shaping the plant microbiome and contribute to the filtering of microbes by plants.

Straw incorporation is known as an environmentally friendly agricultural practice that can effectively enhance soil nutrient contents and crop yields; its potential to suppress soil-borne disease has also been reported in recent years. Here, we perform a field experiment for two consecutive years (2017–2018) to evaluate the effectiveness of maize (Zea mays), rice (Oryzae sativa L.) and wheat (Triticum aestivum L.) straws incorporation in alleviating Chinese cabbage (Brassica rapa L. ssp. pekinensis) clubroot disease caused by Plasmodiophora brassicae Woronin. Microbial composition in Chinese cabbage rhizosphere and soil P. brassicae abundance were estimated by high-throughput amplicon sequencing and quantitative polymerase chain reaction (qPCR). Results showed that, during the two-year field experimental cycle, all three straw amendments promoted Chinese cabbage plant growth, inhibited clubroot disease and increased the alpha diversity of the bacterial community in Chinese cabbage rhizosphere. Rice and wheat straws also increased the alpha diversity of the fungal community. These straws diversified the composition of the Chinese cabbage rhizosphere microbial community. All three straws promoted Cryptococcus carnescens; both rice and wheat straws stimulated Lysobacter sp.; maize straw boosted Sphingomonas sp. and wheat straw increased Talaromyces sp. These microbial taxa are either considered to have positive influences on plant growth or potential biocontrol effects. In addition, straw amendments also increased soil pH, electrical conductivity, available nitrogen and available potassium contents in both years of the field experiment. Taken together, we concluded that these three gramineous straw amendments ameliorated Chinese cabbage rhizosphere microorganisms, inhibited clubroot disease and promoted the growth of Chinese cabbage, and that rice straw worked best amongst the three. This study could potentially provide a new tactic of massive grain crop straw utilization and a direction in dealing with clubroot disease.
