Effects of growth factors (EGF, PDGF‐BB and TGF‐β1) on cultured equine epithelial cells and keratocytes: implications for wound healing

Abstract: Methods were established to maintain epithelial cells and keratocytes in vitro. PDGF-BB and EGF stimulate, while TGF-beta 1 inhibits the proliferation of epithelial cells and keratocytes. These growth factors may play a role in maintenance and repair of the equine cornea.

“…In diabetic patients, a minor skin wound often leads to chronic, nonhealing ulcers and ultimately results in infection, gangrene, even amputation [3,4]. Many experiments have demonstrated that the processes of wound repair were controlled by a wide variety of different growth factors and cytokines, such as PDGF, fibroblast growth factor, epidermal growth factor, insulin-like growth factor, and so on [13][14][15][16][17]. Their expression dynamics showed characteristic temporal and spatial regulation, and changes in the expression pattern of growth factors were associated with impaired wound healing.…”

“…HGF probably modulates the corneal epithelial cell proliferation, motility and differentiation and plays an important role in the homeostasis in corneal epithelium [4,21,44]. The TGF-β family are prototypical fibrogenic cytokines responsible for fibroblast activation in wound healing [16,43]. FGF stimulates corneal stromal wound healing either by direct stimulation or through activation of TGF-β-mediated cell proliferation of fibroblasts [17,30].…”

“…Zwei wichtige GF für die Gewebsregeneration stellen PDGF-BB (plateletderived growth factor BB) und TGF-b 1 (transforming growth factor b 1 ) dar. Während PDGF-BB vor allem ein sehr guter Chemoattraktor ist und die Zellproliferation stimuliert (Anitua et al 2004), ist TGF-b 1 insbesondere für die Steigerung der Kollagensynthese verantwortlich (Haber et al 2003).…”
