Effects of hyperthermia on the cytoskeleton and focal adhesion proteins in a human thyroid carcinoma cell line

Abstract: Hyperthermia is reported to act as a sensitizer to chemotherapeutic drugs in the treatment of cancer. Thyroid follicular carcinoma were used to elucidate the effects of hyperthermic treatment (41-43 degrees C) on cell morphology, cytoskeleton, and the focal adhesion complex. The critical temperature that resulted in inhibition of cell proliferation as the cell number in the same area did not increase over a 23 h time course and irreversible changes in cell morphology was 42-43 degrees C. An immunofluorescence … Show more

“…The cytoskeleton is a cellular component that is common to the plasma membrane, nuclear membrane, and cytoplasmic organelles. The three components of the cytoskeleton (i.e., actin filaments, microtubules, and intermediate filaments) work together to enhance structural integrity, impart cell shape, provide cell motility, and position mRNA and protein [31,34], and they play an important role in cell signaling [37], mitosis, and cytokinesis [38]. During the first three cleavage divisions, the mammalian embryo spends the majority of the cell cycle in S and M phases [39].…”

“…Sometimes the terms mild hyperthermia or just hyperthermia is used for temperatures up to 42 °C and extreme hyperthermia for higher temperatures [ 23 ]. At those temperatures, heat alone may cause protein denaturation [ 24 ], damage to the cytoskeleton [ 25 , 26 ], impairment of certain DNA repair processes [ 27 , 28 , 29 , 30 , 31 , 32 ], changes in cell membrane permeability [ 33 , 34 ], and stimulation of the immune system [ 35 , 36 , 37 , 38 ].…”
