Effects of life cycle and leaves location on gene expression and glycoside biosynthesis pathway in Stevia rebaudiana Bertoni

Oral squamous cell carcinoma is one of the most common high malignant tumors. This experiment aimed to investigate whether ethyl acetate extract of peony (Paeonia suffruticosa) seed coat could affect the proliferation and apoptosis of oral squamous carcinoma cells by regulating the miR-424-3p/STAT3/ Survivin pathway. For this purpose, oral squamous cell carcinoma cell CAL27 was cultured in vitro, and cells were treated with ethyl acetate extract of peony seed coat at different concentrations. MTT was used to detect cell proliferation. Flow cytometry was used to detect apoptosis. qRT-PCR was used to detect the expression level of miR-424-3p. The miR-424-3p mimics and anti-miR-424-3p were transfected into CAL27 cells respectively, and the cell proliferation and apoptosis were detected by the above method. Western blot method was used to detect the expression of PCNA, Bcl-2, Bax, p-STAT3 and Survivin protein. Results showed that ethyl acetate extract of peony seed coat could reduce cell proliferation rate and the protein levels of PCNA, Bcl-2, p-STAT3, Survivin and the expression level of miR-424-3p (P<0.05), increase apoptosis rate and the protein level of Bax (P<0.05). After transfection with anti-miR-424-3p, the cell proliferation rate, the protein levels of PCNA and Bcl-2 were significantly reduced (P<0.05), the apoptosis rate and the protein level of Bax were significantly increased (P<0.05), while the effect of miR-424-3p mimics was the opposite. Transfection of miR-424-3p mimics could significantly reduce the regulatory effect of ethyl acetate extract of peony seed coat on CAL27 cell proliferation, apoptosis and STAT3/Survivin pathway. It concluded that ethyl acetate extract of peony seed coat could inhibit the activation of the STAT3/Survivin signaling pathway by down-regulating the expression of miR-424-3p, thereby inhibiting the proliferation of oral squamous carcinoma cells and inducing apoptosis.

Acting as a really common cancer in the world, bladder cancer has taken many people's life away. MiRNAs and mRNA have been reported can regulate the expression of cancers. In this study, the role of RAB2A and miR-381-3p was fully studied in bladder cancer. qRT-PCR assay probe the expression of RAB2A and miR-381-3p in bladder cancer cells. Meanwhile, colony formation assay, EdU assay, flow cytometry analysis, JC-1 assay and western blot assay were implemented to detect the progression of bladder cancer cells. Silenced RAB2A could reduce the cell proliferation of bladder cancer, and activate the apoptosis.Meanwhile, miR-381-3p could bind to RAB2A in bladder cancer cells and overexpressed miR-381-3p could inhibit the progression of bladder cancer cells. Effects of dendrobium polysaccharides on human brain microvascular endothelial cell injury induced by ox-LDL via regulating the miR-378 expression

Brain microvascular endothelial cells are a key part of the blood-brain barrier. This experiment was set up to investigate the effect and molecular mechanism of Dendrobium polysaccharide on oxidized low-density lipoprotein (ox-LDL)-induced damage to the human brain microvascular endothelial cells.For this purpose, human brain microvascular endothelial cells HBMEC were divided into control group (without any treatment), ox-LDL group (50 μg/mL ox-LDL), Dendrobium polysaccharide low, medium and high concentration group (0.1 μg/L, 0.2 μg/L, 0.4 μg/L Dendrobium polysaccharide+50 μg/mL ox-LDL), ox-LDL+miR-NC group (transfection miR-378 mimic negative control+50 μg/mL ox-LDL), ox-LDL+miR-378 group (transfected miR-378 mimics+50 μg/mL ox-LDL), ox-LDL+DP+anti-miR-NC group (transfected miR-378 inhibitor negative control +0.4 μg/L Dendrobium polysaccharide+50 μg/mL ox-LDL), ox-LDL+DP+anti-miR-378 group (transfected miR-378 inhibitor+0.4 μg/L Dendrobium polysaccharide+50 μg/mL ox-LDL ). The kit was used to detect the levels of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and catalase (CAT); flow cytometry to detect apoptosis; and Western blot to detect B-cell lymph tumor/leukemia-2 (Bcl-2) and Bcl-2 related X (Bax) protein expression; real-time fluorescence quantitative PCR (RT-qPCR) was used to detect the expression of miR-378. Results showed that after treatment with different concentrations of Dendrobium polysaccharides, MDA levels were decreased in ox-LDLinduced human brain microvascular endothelial cells, SOD and CAT activities were increased, apoptosis rate was decreased, Bcl-2 expression was increased, Bax expression was decreased, miR-378 expression was increased, in a dose-dependent manner (P<0.05). Overexpression of miR-378 inhibits ox-LDL-induced oxidative stress and apoptosis in human brain microvascular endothelial cells. Inhibition of miR-378 expression reversed the effect of Dendrobium polysaccharide on ox-LDL-induced damage to human brain microvascular endothelial cells. Then dendrobium polysaccharide may inhibit ox-LDL-induced oxidative stress and apoptosis in human brain microvascular endothelial cells by up-regulating the expression of miR-378.
