Effects of memantine on the passive avoidance test in young rats

Abstract: Alzheimer’s disease (AD) is a long-lasting progressive neurodegenerative disease that degrades memory and cognitive function and is often complicated by disorientation and other psychiatric syndromes. At present, to improve the condition of patients with AD, for their treatment, use the drug memantine. The drug is a noncompetitive antagonist of NMDA glutamate receptors in the brain. The present experiments aimed to test the influence of memantine on the memory processes in rats. We used the passive avoidance t… Show more

“…The results of the present study indicate an increase in post-shock latency in the treatment group of baicalein with memantine at a 20 + 10 mg/kg dose. The improvement in post-shock latency was also observed in the memantine treatment group by Gorbachenko and Lukyanetz (2020) and the baicalein-treated group by Wong et al (2014). Those findings are in line with our study results.…”

“…Memantine has also shown procognitive effects in preclinical AD models. In rat experiments, memantine prevented short-term memory deficits caused by Aβ1-40, improved long-term memory consolidation impaired by soluble beta-amyloid, enhanced spatial memory in animals injected with Aβ1-42 oligomers, and improved performance in other behavioral memory tests (Kruchenko et al, 2014;Tyshchenko and Lukyanetz, 2017;Gorbachenko and Lukyanetz, 2020). The effect of memantine on the viability status of hippocampal neurons treated by Aβ1-42.…”

“…In this study, memantine in week 8 significantly reduced the frequency of avoidance symptom compared to baseline but had no significant effect on other symptoms (Intrusion and Hyperarousal). A preferential reduction in the excitatory impulse to inhibitory neurons in the cortical circuits and subsequent changes in the balance between excitation and inhibition are two mechanisms suggested to explain the therapeutic benefits of memantine ( 31 ).…”
