Effects of modified amphotericin in experimental systemic candidiasis

Abstract: Lysosomotropic composition of dialdehyde dextran and amphotericin B had a greater therapeutic effect in mice with systemic candidiasis compared to free amphotericin B. This composition normalized glucocorticoid function of the adrenal glands and decreased the severity of liver destruction at late terms of granulomatous inflammation.

“…This also attest to higher therapeutic efficiency of CA compared to AmB, which agree with previous reports [8,9]. The efficiency of both forms of the antibiotic in the treatment of candidiasis in CBA mice was lower than in C57Bl/6 mice opposite to CBA mice by some functional parameters of the pituitary-adrenal and mononuclear phagocyte systems [2,3,5].…”

“…The efficiency of both forms of the antibiotic in the treatment of candidiasis in CBA mice was lower than in C57Bl/6 mice opposite to CBA mice by some functional parameters of the pituitary-adrenal and mononuclear phagocyte systems [2,3,5]. For instance, no infiltrates and granulomas were found in the liver of C57Bl/6 on day 56 of AmB treatment, while in animals treated with CA this effect was noted as soon as on day 28 [9].…”

“…The toxic effects of these metabolites are manifested in destructive processes in various organs [11,13] and not associated with the presence and number of granulomas. They are probably related Nadeev, et al to the influence of fungal metabolic products and manifested in modulation of steroidogenesis [10,12]. In our experiments, AmB did not exhibit nephrotoxicity.…”

“…The fact that it was water soluble makes it potentially useful for intravasacular injection. AmB has been conjugated to other hydrophilic polymers and natural polysaccharides including arabinogalactan [14], dextran [15, 22], gum arabic [16] and PEG [23]. These conjugates have been shown to reduce the toxicity of AmB in mice [14, 16, 22, 23].…”
