Effects of Olibanum extracts on the collagenase activity and procollagen synthesis in Hs68 human fibroblasts and tyrosinase activity

Abstract: Abstract. This study was designed to investigate the collagen metabolism and tyrosinase activity of Olibanum extracts (OE). The traditional effects are known to move blood, calm pain, solve oedema, and promote tissue regeneration. The effect of OE on type I procollagen production and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts Hs68 after ultraviolet B (UVB, 312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment o… Show more

“…It induces activator protein-1 (AP-1), a transcription factor that promotes collagen breakdown by up regulating enzymes called matrix metalloproteinases (MMPs). Overexpression of matrix metalloproteinase (MMP-1, MMP-3 and MMP-9) leads to elevated levels of degraded collagen (Leem, 2015). Also, UV radiation causes downregulation of transforming growth factor ␤ (TGF-␤), a cytokine that promotes pro-collagen production thus causing reduced collagen synthesis (Fig.…”

“…Skin aging is caused by an intrinsic or natural mechanism affecting the skin and other body organs related to hormonal changes occurring with age, and by extrinsic mechanisms associated with ultraviolet radiation exposure, which causes free radical species generation [1]. Reactive oxygen species (ROS) are responsible for inducing the activator protein-1 (AP-1), a transcription factor that promotes collagen and elastin breakdown by up regulation of the matrix metalloproteinases (MMPs) [2]. This environmental stress generated from ROS and reactive nitrogen species (RNS), combined with increased secretion of inflammatory mediators and enzyme expression (e.g., collagenase and elastase), causes inflammation and decreases the tensile strength and elasticity of the skin [3].…”

“…However, the FoxO is rendered inactive by the AKT pathway, which prevents the cell's production of antioxidant enzymes. Activator protein-1 (AP-1) and NF-κB are both nuclear proteins that MAPK regulates, and the expression of MMP is caused by AP-1 stimulation [46]. MMPs are a group of extracellular proteinases that include zinc that breaks down extracellular substances, including collagen and elastic fibers, which results in the development of wrinkles [46,47].…”
