“…Our studies were conducted using charcoal stripped and dialyzed serum that does not support VDR dependent transcription in the absence of an exogenous source of ligand. 85 It is known that VDR can bind other ligands such as curcumin 86 and lithocholic acid, 87 the latter which is a toxic bile acid that activates VDRdependent transcription of the CYP3A detoxifying gene that is independent of VD3. However, it remains formally possible that trace amounts of VD3 in our medium is sufficient to facilitate VDR dependent repair functions.…”

“…While many reports indicate '' > 99%'' of T 3 and T 4 are removed via standard stripping protocols, some variability exists in the degree of T 3 and T 4 removal achieved by the various procedures. This variability may arise both from factors related to the type of serum and from the method of stripping (257,258). Furthermore, current chemical stripping methods do not allow for the depletion of thyroid hormone without also depleting other circulating factors.…”

“…Furthermore, current chemical stripping methods do not allow for the depletion of thyroid hormone without also depleting other circulating factors. Many small molecules such as growth factors or hormones (including sex steroids, adrenal steroids, and vitamin D) may also be removed during stripping, and thus the biologic changes seen over time cannot be described as representing isolated hypothyroidism (257). Experimental design should compare groups with thyroid hormone replaced (versus vehicle replaced) to enable specific attribution of the results to thyroid hormone deficiency.…”

“…Further, a suppressive effect of steroid-stripped media on antigen-induced RBL degranulation is documented here. Thus, it is clear that at least some of the many hormones and other small biomolecules that are depleted in the charcoal stripping process (Cao et al, 2009) are important for robust mast cell functioning.…”
