Effects of rikkunshito on renal fibrosis and inflammation in angiotensin II-infused mice

Abstract: The underlying pathogenesis of chronic kidney disease involves an activated renin-angiotensin system and systemic inflammation which ultimately develop renal injury. Rikkunshito (RKT) has been reported to exert anti-fibrotic and anti-inflammatory effects through enhancement of ghrelin signaling pathway. In this study, we investigated the effects of RKT on renal fibrosis and inflammation in angiotensin II (Ang II)-induced renal injury model. Ang II-infused mice exhibited hypertension, cardiac hypertrophy, incre… Show more

“…Fujitsuka et al reported that RKT administration in a senescent mouse model activates SIRT1, inhibits inflammation and apoptosis associated with aging, and prolongs life span 37 . In addition, we have very recently reported that Ang II-induced decrease in renal SIRT1 expression was restored by RKT, concomitant with a suppression of renal inflammation in Ang II-infused mice 11 . In the present study, renal SIRT1 protein expression was antagonistically increased in response to UUO in mice.…”

“…Immunoblot analysis was performed as described previously 11,22 . Briefly, total protein was extracted from kidney tissues with sodium dodecyl sulfate-containing sample buffer.…”

“…The expression levels of pro-inflammatory factors ( 32 , 33 ) were also detected in liver tissues and tumor tissues from mice in the STZ-HFD group, and it was found that the expression levels of IL-6, TNF-α, GPC3 and TGF-β were upregulated in the tumor tissues. Notably, GPC3, a heparan sulfate proteoglycan binding to the cell membrane by glycosylphosphatidylinositol, is hardly expressed in adults, except in the placenta.…”

“…Systolic blood pressure and heart rate were measured using the tail-cuff method (BP-Monitor MK-2000; Muromachi Kikai Co., Tokyo, Japan), as previously described 20,21 . All measurements were performed between 9:00-14:00 h. At least 10 measurements were performed in each mouse and the mean value was used for analysis.…”
