Synchrony of energy and nitrogen release in rumen has been proposed to maximize ruminal microbial fermentation. However, the information regarding bacterial community composition and its metabolism under a higher or lower degree of synchronization is limited. In our study, a 0 to 6 h post-feeding infusion (first half infusion, FHI), 6 to 12 h post-feeding infusion (second half infusion, SHI), and 0 to 12 h post-feeding infusion (continuous infusion, CI) of maltodextrin were used to simulate varying degrees of synchronization of energy and nitrogen release in a rumen simulation system. In addition, the bacterial community, metabolite, enzyme activity, and microbial protein synthesis (MPS) were evaluated. Compared with the FHI and CI, the relative abundance of Fibrobacter, Ruminobacter, BF311, and CF231 decreased in the SHI, but that of Klebsiella and Succinivibrio increased in the SHI. The NH3-N and branched-chain volatile fatty acids were significantly higher, but propionate content and activities of glutamate dehydrogenase (GDH) and alanine dehydrogenase were significantly lower in the SHI than those in the FHI and CI. The SHI had lower MPS and less efficiency of MPS than the FHI and CI, which indicated that the SHI had a lower degree of synchronization. Correlation analysis showed that MPS was positively related to GDH activity and relative abundance of Fibrobacter but negatively related to NH3-N and relative abundance of Klebsiella. Therefore, a higher degree of synchronization of energy and nitrogen release increased MPS partly via influencing the bacterial community, metabolism, and enzyme activities of ammonia assimilation in the in vitro fermenters.

Nos sistemas de produção de ruminantes a alimentação pode responder por 60 a 70% dos custos da atividade, sendo assim, a avaliação nutricional dos alimentos e a formulação de dietas balanceadas, que atendam às exigências dos animais representam um importante desafio na produção pecuária. Diversos métodos têm sido usados para avaliar a modulação da fermentação ruminal e estimar as características da degradação de alimentos e dietas destinadas à alimentação de ruminantes. Esses métodos envolvem a medição do desaparecimento do substrato durante a incubação no rúmen in situ, no líquido ruminal tamponado (in vitro) e são principalmente medidas de ponto final. Além disso, é possível avaliar a cinética da degradação através da medição das diferenças nos resíduos em diferentes tempos de incubação. A avaliação do valor nutricional de alimentos e dietas também é possível ser feita por meio da utilização de técnicas que medem a produção de gás. Essas técnicas medem o aparecimento de produtos da fermentação ruminal, tais como os gases dióxido de carbono (CO2) e metano (CH4), ácidos graxos de cadeia curta, amônia e a biomassa microbiana. Objetivou-se com essa revisão descrever os principais métodos in situ e in vitro utilizados para avaliar alimentos para ruminantes.

Pearl millet crop has been increasingly growing in Brazilian Savanna and it is already being used as cover crop between annual crops. The plant has great forage potential, besides being a nutrient recycling plant due to its peculiar root system. This study was developed in order to assess the pearl millet nutritional value when submitted to nitrogen fertilizer. It was evaluated the effect of nitrogen fertilizer (ammonium sulfate nitrate) treated with the nitrification inhibitor 3,4-dimethyl pyrazole phosphate (DMPP) on the ruminal degradability of two pearl millets' cultivars, under four nitrogen fertilization levels (0, 45, 90 and 180 kg ha ) and pre-cutting heights (0.70, 0.80 and 0.90 m). The experimental design was a randomized block design in a factorial 3 × 4 (3 cutting heights × 4 nitrogen doses) with three replications. Data were submitted to analysis of variance and means were compared by Tukey test at 5% probability. The DMPP treated Nitrogen, in high doses, increased the dry matter, crude protein and neutral detergent insoluble fiber degradability in pearl millet handled at 0.90 m. The combination of fertilization with 45 or 90 kg ha -1 of nitrogen treated with DMPP, with the management of millet at 0.70 or 0.80 m did not favored the forage nutritional quality, indicating that in these treatments, the ratio between the availability of nitrogen in ammonium and nitrate forms may have been detrimental to the plants.
