The increasing prevalence of environmental pollutants such as pesticides is a major global problem that affects living organisms. Exposure to environmental pollutants remains a major source of health risk throughout the world. The potential health benefits of various medicinal plants and natural products in relation to protect various diseases are currently receiving considerable attention. A current approach is to develop a new biological compound from natural products that inhibits pain. Ethnopharmacological surveys have been found to be one of the most reliable tools for the discovery of the natural and semi-synthetic drug. The present study was performed to investigate the hematological and biochemical changes induced by carbendazim (CBZ) and the potential protective effect of seeds extracts of Nigella sativa (NSSE) and Foeniculum vulgare (FVSE) against CBZ toxicity in male mice. Mice were distributed into 6 groups. Mice of group 1 were served as control. Group 2 was exposed to CBZ. Group 3 was supplemented with NSSE and exposed to CBZ. Group 4 was treated with FVSE and CBZ. Normal mice of group 5 and 6 were subjected to NSSE and FVSE respectively. Body weight gain was significantly decreased in mice of group 2. In mice of group 2, significant declines of RBC, HB, Hct, WBC, total protein, FSH, LH, testosterone, T4, T3, CAT and SOD were observed. Moreover, the levels of ALT, AST, ALP, total bilirubin, creatinine, BUN, uric acid, glucose, cholesterol, CK, LDH, MDA and GSH were significantly enhanced. Treatment with NSSE and FVSE showed attenuation effects against CBZ induced hematological and biochemical changes. The results suggest that the attenuation effects of NSSE and FVSE attributed to their antioxidant properties.

Humans are exposed to pyrethroid‐based pesticides through agricultural produce. In this study, male Wistar rats were orally treated for 9 to 12 months with a mixture of pyrethroids that is equivalent to one‐fifth (high dose; HD) or one‐twenty fifth (low dose; LD) of the amount of pyrethroids present in the cereals and rice consumed by an average Indian. In rats treated for 9 months, the spermatogenesis‐associated genes Abp, Ar, Cd9, Dax1, Dazap1, Ddx3y, Gdnf, Gfra1, Grth, Inhb, Ovol1, P1, Plzf, Pygo2, Scf, Tgfb1, Tp1, Tp2, and Vim1 were downregulated in both LD and HD groups. In rats treated for 12 months Gdnf, Hsf2, Inhb, Tgfb1, Thy1, and Ybx2 expression was downregulated in both LD and HD groups. Steroidogenesis‐associated genes 17‐β‐Hsd, Gata4, Hmgcr, Hmgcs1, Pde4b, and Tspo gene expression were reduced in both LD‐ and HD‐treated groups treated for 9 months. In 12‐month‐treated rats, Creb1 expression decreased in both LD and HD groups. The epigenetic reprogramming‐associated genes, Dnmt1, Dnmt3a, Dnmt3b, Hdac10, Hp1bp3, Kat3a Kat3b, Mch2ta, Ncoa7, and Sirt1 were downregulated in both HD and LD groups of 9‐months‐treated rats. In rats treated for 12 months, Hdac10, Mch2ta, Ncoa7, and Sirt1 messenger RNA levels decreased in both the HD and LD groups. Thus, we demonstrate that long‐term exposure to a mixture of pyrethroids caused aberrations in the transcriptome of factors involved in sperm production and development.

Exposure to organophosphorus insecticides causes several health problems to animals and humans. Red beetroot (RBR) is rich in antioxidant ingredients and possesses a promising hepatoprotective activity. This study evaluated the potential of RBR extract to prevent chlorpyrifos (CPF)-induced liver injury, with an emphasis on oxidative stress, inflammation and apoptosis. Rats received 10 mg/kg CPF and were treated with 300 mg/kg RBR extract for 28 days. CPF caused liver injury evidenced by elevated serum levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and bilirubin, along with several histological alterations. Hepatic lipid peroxidation (LPO) and nitric oxide (NO) levels, as well as inducible nitric oxide synthase (iNOS) and pro-inflammatory cytokines were increased in CPF-intoxicated rats. RBR prevented CPF-induced histological alterations, and ameliorated liver function, LPO, NO, iNOS and pro-inflammatory cytokines. RBR boosted glutathione and antioxidant enzymes, and increased Nrf2 expression. In addition, RBR diminished Bax and caspase-3, and increased Bcl-2 expression. In conclusion, RBR prevented CPF-induced liver injury via attenuation of oxidative stress, inflammation and apoptosis. RBR enhanced antioxidant defenses, suggesting that it could be used as a potential therapeutic intervention to minimize CPF hepatotoxicity.
