ABSTRACT-The effects of chromium compounds on 3-O-methylglucose (3-0-MG) transport were studied in isolated rat adipocytes. Sodium chromate significantly stimulated 3-0-MG uptake into adipocytes in a dose-dependent manner without altering the equilibrium space of 3-0-MG in adipocytes. The stimulatory effect reached the maximum at 300 pM, and the effect was 60 -70070 of the maximal insulin effect that was obtained with 20 nM insulin. The chromate concentration achieving a half-maximal effect was estimated at 50 pM. The effect of the combination of 1 mM chromate and 20 nM insulin was equipotent to that of 20 nM insulin alone, which showed that these two effects were not additive. The stimulatory effects of 1 mM chromate and 20 nM insulin were entirely abolished in adipocytes deprived of ATP, which indicated that these effects were completely ATP-dependent.Judging from experiments using various chromium compounds, Cr042-was responsible for the insulinomimetic action. These results indicate that the action of Cr042-is exerted through a mechanism analogous to that of insulin action, and that Cr042-is a novel and useful tool for studying issues involved in insulin actions.

This paper gives insight into chemical speciation. After a brief introduction, the methods available to study solution state of metal ions and the importance of speciation in biology are discussed through different examples. Speciation of toxic Al III , beneficial Cu II and Ca II and potentially medicinal Pd II and VO IV in various biofluids and tissues is discussed in order to describe the solution state of these metal ions in an organism. The relevance of ternary complex formation with endogenous biomolecules is emphasised. The importance of the biospeciation of Al III ion in its transport and involvement in neurological disorders, of the actual chemical form of the insulin-mimetic VO IV complexes in its biological occurrence and Ca II in mineralization/demineralization processes is discussed. The role of time, i.e., the kinetic aspects of biospeciation, is discussed in the case of the transport of Al III and Cu II and some Pd II complexes (models of the anticancer Pt II complexes) in blood serum.

Vanadate (sodium orthovanadate), an inhibitor of phosphotyrosine phosphatases (PTPs), mimics many of the metabolic actions of insulin in vitro and in vivo. The potential of vanadate to stimulate glucose transport independent of the early steps in insulin signaling prompted us to test its effectiveness in an in vitro model of insulin resistance. In primary rat adipocytes cultured for 18 h in the presence of high glucose (15 mM) and insulin (10 ؊7 M), sensitivity to insulin-stimulated glucose transport was decreased. In contrast, there was a paradoxical enhanced sensitivity to vanadate of the insulinresistant cells (EC 50 for control, 325 ؎ 7.5 M; EC 50 for insulin-resistant, 171 ؎ 32 M; p < 0.002). Enhanced sensitivity was also present for vanadate stimulation of insulin receptor kinase activity and autophosphorylation and Akt/protein kinase B Ser-473 phosphorylation consistent with more effective PTP inhibition in the resistant cells. Investigation of this phenomenon revealed that 1) depletion of GSH with buthionine sulfoximine reproduced the enhanced sensitivity to vanadate while preincubation of resistant cells with N-acetylcysteine (NAC) prevented it, 2) intracellular GSH was decreased in resistant cells and normalized by NAC, 3) exposure to high glucose and insulin induced an increase in reactive oxygen species, which was prevented by NAC, 4) EPR (electron paramagnetic resonance) spectroscopy showed a decreased amount of vanadyl (؉4) in resistant and buthionine sulfoximine-treated cells, which correlated with decreased GSH and increased vanadate sensitivity, while total vanadium uptake was not altered, and 5) inhibition of recombinant PTP1B in vitro was more sensitive to vanadate (؉5) than vanadyl (؉4). In conclusion, the parodoxical increased sensitivity to vanadate in hyperglycemia-induced insulin resistant adipocytes is due to oxidative stress and decreased reduction of vanadate (؉5) to vanadyl (؉4). Thus, sensitivity of PTP inhibition and glucose transport to vanadate is regulated by cellular redox state.
