Efficacy and tolerability of amorolfine 5% nail lacquer in combination with systemic antifungal agents for onychomycosis: A meta-analysis and systematic review

Abstract: The efficacy and safety of amorolfine 5% nail lacquer in combination with systemic antifungal agents in the treatment of the onychomycosis were evaluated. According to our meta-analysis, combination treatment of amorolfine 5% nail lacquer and systemic antifungals can result in higher percentage of complete clearance of onychomycosis. It showed that the experimental combination group was more effective than monotherapy of the systemic antifungals [OR (odds ratio) = 1.97, 95%CI (95% confidence interval) = 1.44-2… Show more

“…Topical antifungal therapy is a therapeutic option when oral antifungal agents are contraindicated or cannot be tolerated [ 1 , 2 , 44 , 45 , 88 ]. Topical antifungals may also be used in combination with oral antifungal therapy as an adjunctive therapy to increase the cure rate due to synergistic antifungal action of the drugs [ 32 , 88 , 105 , 113 ]. Because the rate of nail growth is faster and the nail plate is thinner in children, children are more likely to respond to topical antifungal therapy better than adults [ 17 , 53 , 88 , 114 , 115 ].…”

“…Feng et al 45 performed a meta‐analysis on the efficacy of amorolfine 5% nail lacquer in combination with systemic antifungal agents. Accordingly, amorolfine combination treatment results in a significantly higher rate of complete clearance of onychomycosis than systemic monotherapy with itraconazole or terbinafine (odds ratio = 1.97; 95% confidence interval = 1.44‐2.69).…”

“…Classical treatments with azoles or terbinafine are mostly effective for DMPs . Once an oral antifungal drug is administered, DMPs, which usually respond to oral antifungal monotherapy, are nearly wholly eradicated.…”
