Efficacy of ?-diketonato complexes of titanium, zirconium, and hafnium against chemically induced autochthonous colonic tumors in rats

Abstract: Bis-beta-diketonato complexes of titanium, zirconium, and hafnium were tested against autochthonous colorectal tumors in rats. The model was found to reflect the clinical situation most closely. Of the compounds tested, budotitane was the most effective in terms of decrease in tumor weight and number and in increasing the lifespan of the treated animals. The therapeutic efficiency was superior to that of 5-fluorouracil, which so far has been the drug with the best activity in patients suffering from colon canc… Show more

“…β‐Diketonate ligands have also been used to deposit WO 3 and CuO . There are also a wide variety of β‐diketonate ligands known for metals including: Ti, Hf, Ta, Sr, Ca, Mg, and Ba, therefore a wide range of metal oxide materials are accessible via such precursors.…”

“…6). The complexes dichlorobis(1-phenylbutane-l,3-dionato)titanium(IV) [TiIV(bzac)2C12] and diethoxybis(1-phenylbutane-l,3-dionato)titanium (IV) [TilV(bzac)2(OEt)2] (budotitane) were the most active compounds (Keppler and Schm~ihl 1986) against the mentioned animal tumours and the chemically-induced autochthonous colon adenocarcinoma in rats (Bischoff et al 1987). “…These compounds had significant antitumour activity against the animal tumours Ehrlich ascites tumour, Stockholm ascitic tumour, sarcoma 180 ascitic tumour, and the solid tumour models Walker 256 carcinosarcoma and MAC 15A colon carcinoma, whereas the leukemias P388 and L1210 responded only marginally (Keppler et al 1985(Keppler et al , 1991. The analogous zirconium and hafnium compounds were less effective than both titanium compounds (Bischoff et al 1987;Keppler et al 1991). “…[15][16][17] Although much effort was undertaken to identify completely new titanium complexes that show antitumor properties, nearly all investigated complexes were derivatives of either titanocene dichloride ([TiA C H T U N G T R E N N U N G (Cl 2 )A C H T U N G T R E N N U N G (Cp 2 )]) [18][19][20][21][22] or budotitane ([TiA C H T U N G T R E N N U N G (bzac) 2 -A C H T U N G T R E N N U N G (OEt) 2 ]; Hbzac = phenylbutane-1,3-dione). [23][24][25] One characteristic of the above-mentioned complexes is their very fast rate of hydrolysis that leads to unidentified products. In an aqueous environment, the loss of the labile groups occurs within seconds, whereas the more inert lig-A C H T U N G T R E N N U N G ands hydrolyze on a timescale of hours or days.…”
