Efficiency of Tris-Based Extender Steridyl for Semen Cryopreservation in Stallions

Abstract: The fertilizing ability of stallion sperm after freezing is lower than in other species. The search for the optimal extender, combination of extenders, and the freezing protocol is relevant. The aim of this study was to compare lactose-chelate-citrate-yolk (LCCY) extender, usually used in Russia, and Steridyl® (Minitube) for freezing sperm of stallions. Steridyl is a concentrated extender medium for freezing ruminant semen. It already contains sterilized egg yolk. Semen was collected from nine stallions, aged … Show more

“…The compositions of extenders are important for semen quality and survival, especially after the freezing–thawing process. Semen extenders have usually been composed of electrolytes (Tris, citric acid), sugars (glucose, fructose), cryoprotectants (egg yolk, glycerol) and antibiotics for several years for semen cryopreservation (Gangwar et al, 2018; Nikitkina et al, 2020). Besides these compounds, antioxidants are very important for mammalian semen freezing.…”

“…Sperm was diluted at 1:3 (v/v) ratio with lactose-chelate-citrate-yolk (LCCY) medium containing 3.5% glycerin and frozen according to a standard technology (standard operating procedure) used at the ARRIHB and described in detail elsewhere [11,[26][27][28]. Briefly, four-cornered aluminum tubes were used to package the diluted semen.…”

“…Andromed ® contains soy lecithin and tris-buffer, while Optixcell ® contains synthetic lecithin (liposome) (Swelum et al 2019). Furthermore, Steridyl ® is a commercial diluent based on tris and sterile egg yolk (Nikitkina et al 2020). Andromed ® composed of buffer (tris-(hydroxymethyl)-aminomethane, citric acid), sugar (fructose), antioxidants, glycerol, water, and four kinds of antibiotics (tylosin tartrate, gentamycin sulfate, spectinomycin, and lincomycin).…”
