Efficient Endocytic Uptake and Maturation in Drosophila Oocytes Requires Dynamitin/p50

Abstract: Dynactin is a multi-subunit complex that functions as a regulator of the Dynein motor. A central component of this complex is Dynamitin/p50 (Dmn). Dmn is required for endosome motility in mammalian cell lines. However, the extent to which Dmn participates in the sorting of cargo via the endosomal system is unknown. In this study, we examined the endocytic role of Dmn using the Drosophila melanogaster oocyte as a model. Yolk proteins are internalized into the oocyte via clathrin-mediated endocytosis, trafficked… Show more

“…Moreover, signal transduction as well as protein trafficking pathways in Leishmania are not well characterized; therefore, the identification of effector molecules is more challenging in parasites. However, we have found some LdRab5 interactors like dynein, kinesin, and tubulin that are shown to regulate endocytosis and vesicular trafficking (59,60). Moreover, Rab5 is also reported to transport cargo to the nucleus (61); thus, interaction with a nuclear transport factor is an interesting observation.…”

“…Ovaries were dissected as previously described (Liu et al 2015) and fixed in either 8% formaldehyde for 10 min (for the a-tubulin staining experiment) or 4% formaldehyde for 20 min (for the remaining experiments). The fixative was diluted in PBS.…”

“…CME is fundamental for a wide range of cellular processes, including nutrient uptake, synaptic vesicle recycling, signal transduction regulation, and receptor internalization . For nonmammalian species, such as Drosophila , mosquitos, and chickens, CME is a critical process utilized by oocytes to import yolk protein precursors during oocyte growth. In addition, receptor endocytic trafficking is vital for oocyte meiotic maturation in Caenorhabditis elegans , and blocking CME negatively modulates Xenopus oocyte maturation .…”
