The orexigenic peptide ghrelin has been shown to have prokinetic activity in the gastrointestinal (GI) system of several species, including humans. In this series of experiments, we have evaluated the prokinetic activity of novel, small-molecule ghrelin receptor (GhrR) agonists after parenteral and peroral dosing in mice and rats. Gastric emptying, small intestinal transport, and fecal output were determined after intraperitoneal and intracerebroventricular dosing of GhrR agonists, using ghrelin as a positive control. These same parameters were evaluated after oral gavage dosing of the synthetic agonists. Regardless of dose route, GhrR agonist treatment increased gastric emptying, small intestinal transit, and fecal output. However, fecal output was only increased by GhrR agonist treatment if mice were able to feed during the stimulatory period. Thus, GhrR agonists can stimulate upper GI motility, and the orexigenic action of the compounds can indirectly contribute to prokinetic activity along the entire GI tract. The orexigenic and prokinetic effects of either ghrelin or small-molecule GhrR agonists were selective for the GhrR because they were absent when evaluated in GhrR knockout mice. We next evaluated the efficacy of the synthetic GhrR agonists dosed in a model of opiate-induced bowel dysfunction induced by a single injection of morphine. Oral dosing of a GhrR agonist normalized GI motility in opiate-induced dysmotility. These data demonstrate the potential utility of GhrR agonists for treating gastrointestinal hypomotility disorders.

In our continuing efforts to find new antiherpetic agents from plants, an extract prepared from the stems of Carissa spinarum L. was found to possess appreciable activity against herpes simplex viruses (HSV I and II). A chemical study of this plant was then initiated, and this led to the isolation of 12 compounds, including a coumarin, two cardiac glycosides and nine lignans. These isolated compounds were evaluated for several biological activities, including antiherpetic, cytotoxic, antioxidant and antibacterial effects. The cardiac glycoside evomonoside was found to be the only antiherpetic principle, showing moderate activity against herpes simplex virus types I and II in the inactivation method. The lignans (-)-carinol, (-)-carissanol and (-)-nortrachelogenin exhibited cytotoxicity against breast (MCF7) and lung (A549) cancer cells. Moderate anti-DPPH free radical activity was observed for all the lignans. None of the isolates showed antibacterial activity.

The present study was conducted to investigate in vitro and in vivo antibacterial activity of the root bark extracts of Anogeissus leiocarpa (DC) Guill. & Perr, in Escherichia coli extended-spectrum βlactamases (ESBL)-induced diarrhea in rat. The antibacterial activity was performed in vitro by determining the inhibition zone using standard agar diffusion method as well as in vivo on E. coli infected Wistar rat model. Both minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were carried through microdilution method. Results obtained in this study indicated that ethanolic and acetatic extracts were only active on bacteria presenting an inhibition zone range from 8 to 16 mm. The MIC observed in agar slant tubes ranged from 6.25 to 50 mg/ml. The ethanolic fraction of A. leiocarpa (ETHA) showed the highest in vitro antibacterial activity against strains with MICs ranging from 6.25 to 12.5 mg/ml and MBCs ranging from 12.5 to 25 mg/ml. In vivo, after infection, diarrhea increased faeces frequency, weight and volume faeces and bacterial faeces load to a maximum on the 2nd day after infection (P < 0.01). ETHA normalized the appearance, weight, volume and water content of faeces. To all doses, like ciprofloxacin it reduced significantly (P < 0.05) the bacterial growth compared to control Lot (infected and untreated). The death rate in diarrheic control Lot was 50% by Day 14. No death was recorded in Lot treated with ciprofloxacin and Lot treated with ethanolic fraction at dose of 2222.22 mg/kg body weight (bw). This study supports the use of A. leiocarpa in the traditional treatment of bacterial infections and offer many perspectives in the search for new molecules against resistant microbial strains.
