Elimination of ornithogalum mosaic virus in the Ornithogalum cv. Rogel through meristem tip culture and chemotherapy

Abstract: Virus-indexed Ornithogalum cv. Rojel plants were produced by eliminating Ornithogalum mosaic virus (OMV) through meristem-tip culture. The best plantlet regeneration was obtained from meristems derived from adventitious buds which developed on leaf explants taken from mother plants at the flowering stage. Acyclovir had no effect as an anti-viral compound on plantlet regeneration or virus elimination. Adenine arabinoside retarded plantlet development at concentrations of 10mg1-1 and higher, while 5.0 mg 1 -I su… Show more

“…Acyclovir had no effect on Potato virus S (Park et al 1994) and on Ornithogalum mosaic potyvirus (Vcelar et al 1992). Caner et al (1985) showed its inhibitory effect on Bean golden mosaic virus infecting Phaseolus lunatus when sprayed on the preinoculated plants.…”

“…Although thermotherapy has been applied to eliminate LSV effectively in L. × elegans (Nesi et al, 2009), there has been no report on the successful elimination of CMV and LMoV using thermotherapy; therefore, it seems that meristematic tip culture and either meristematic tip culture or chemotherapy should be used for LMoV and CMV elimination, respectively, and meristematic tip culture, chemotherapy or thermotherapy can eradicate LSV. Multiplication in vitro, acclimation, and cultivation in a phytotron It has been reported that the virus infection rate increased in acclimated plantlets in many bulbous plants (Phillips, 1990;Vcelar et al, 1992;Xu and Niimi, 1999;Xu et al, 2000). In this study, to check for virus propagation after acclimation, virus-free bulblets produced by meristematic tip culture with DHT were grown in the phytotron and examined by RT-PCR virus detection methods.…”

“…This necessitates the production of certified healthy virus-free propagation material for establishing new orchards. Shoot tip grafting (STG), shoot tip/ meristem culture, thermotherapy, and chemotherapy were used separately or in combination to eliminate viruses from the plants (Vcelar et al 1992;Fletcher et al 1998;Manganaris et al 2003;Nascimento et al 2003;Valero et al 2003;Ram et al 2005;Verma et al 2005).…”
