EMT Transcription Factors Are Involved in the Altered Cell Adhesion under Simulated Microgravity Effect or Overloading by Regulation of E-cadherin

Abstract: In order to study the effect of stress changes on cell adhesion, HUVEC, and MCF-7 cells were treated with simulated microgravity effect (SMG) and overloading (OL). Methods: Rotating Wall Vessel (2D-RWVS) bioreactor was used to create different culture conditions. In addition, the alteration of cell adhesion states, adhesion proteins, and relating factors of adhesion molecules under these two conditions were detected using cell adhesion assay, immunofluorescence, western blot, and qRT-PCR technology. Results: T… Show more

“…The downregulation of β 1 -integrin may inhibit cell adhesion to the ECM. In addition, MCF-7 breast cancer cells exposed to the rotating wall vessel (RWV) showed a decrease in β 1 -integrin protein [34], supporting the recent finding that focal adhesion is reduced in space and under s-µg conditions [35].…”

“…In breast cancer, Wnt3 overexpression can promote EMT-like phenotype through activating the Wnt/β-catenin signaling pathway [ 47 ]. EMT involves the down-regulation of E-cadherin and plays a vital role during the early stage of cancer metastasis [ 32 , 34 , 48 ]. EMT may result in the loss of intercellular connections of cancer cells, upregulation of contractile cytoskeleton components and remove the benign primary on account of epithelium; these changes make EMT a key factor that should not be neglected when studying the development of cancer [ 49 ].…”

“…For example, in tumor cells, the expression of ICAM1 protein and the performance of focal adhesion such as vinculin and cofilin are different between real and ground-based simulated microgravity [82]. The expression of E-cadherin and cell adhesion state in HUVEC and MCF-7 cells showed different under simulated microgravity and overloading condition [110]. In addition, bauer et al used mass spectrometry and mRNA microarray to compare the different effect of microgravity on sialylation of adhesion proteins in FTC-133 thyroid cancer cells, MCF7 cells, and EA-hy926 cells [111].…”
