Enantioselective liquid chromatography–mass spectrometry assay for the determination of ifosfamide and identification of the N-dechloroethylated metabolites of ifosfamide in human plasma

Abstract: The lower limit of detection (LLOD) was 5.00 ng/ml. The inter-and intra-day precision ranged from 3.63 to 15.8 % relative standard deviation (RSD) and 10.1 to 14.3 % RSD, respectively, and the accuracy ranged from 89.2 to 101.5 % of the nominal values. The method was applied to the analysis of plasma samples obtained from a cancer patient who received 3.75 g/m 2 /d dose of (R,S)-ifosfamide as a 96-h continuous infusion.

“…115,116 IFO is a nonionic drug, and nonionic drugs are difficult to intercalate into LDHs. Nie and Hou intercalated IFO into Mg/Al LDHs by the surfactant-assisted method, in which they applied sodium dodecyl sulfate and sodium dodecylbenzenesulfonate.…”

“…21 Oxazophosphorine compounds are chiral molecules (asymmetric phosphorus) administered as a racemate mixture to the patient (the S-enantiomer has more potent anticancer activity). Protein-based columns [187][188][189][190][191][192] or polysaccharide-based columns 193,194 allow resolution of oxazophosphorine enantiomers. Ifosfamide enantiomers and their metabolites in urine and serum can be discriminated by a β-cyclodextrine capillary GC column.…”

“…Ifosfamide contains a chiral centre and is prescribed as a racemic mixture, but the two enantiomers are metabolised by different cytochrome P450 enzymes. It may be advantageous to separate and quantify the two enantiomers to obtain a clear picture of the pharmacokinetics and metabolism of this drug; this can be achieved using SPE with HLB cartridges and chromatography on a Chirabiotic T column [188]. Busulfan is used in preparative regimes for haemopoieitc stem cell transplantation in adults and children.…”
